摘要
目的以自建的人神经母细胞瘤转移瘤细胞(QDDQ-NMl)及原位瘤细胞(ODDQ-NM2)为研究对象探讨KAll/CD82在不同转移潜能人神经母细胞瘤细胞系细胞中的表达情况。方法采用RT-PCR检测两神经母细胞瘤细胞系细胞KAllmRNA的表达;免疫组化方法检测细胞(、D82的表达。结果RT-PCR显示QDDQ-NMl细胞系KAllmRNA的相对含量为0.0748±0.0063,QDDQ-NM2细胞系为0.4576±0.0226,两者比较差异有统计学意义(P〈0.05),免疫组化法检测两细胞系细胞CD82蛋白表达的阳性率分别为30%和75%,两者的差异有统计学意义(P〈0.05)。结论神经母细胞瘤细胞系的细胞高表达KAll/CD82,与人神经母细胞瘤细胞系细胞的体外高转移潜能呈负相关。
Objective To study the expression of KAI1/CD82 in the human neuroblastoma with different metastatic potential. Methods Neuroblastoma NB metastatic tumor (QDDQ-NM) and primary tumor (QDDQ-NM2) cell lines were used in this study. The mRNA expression of KAI1 was determined by reverse transcription-polymerase chain reaction (RT-PCR). The protein level of CD82 was studied by immunocytoehemical staining. Results The mRNA relative expression level of KAI1 in QDD~ NM1 cell lines was 0. 074 8 -+ 0. 006 3, which was significantly higher than that in QDDQ- NM2 cell lines (0. 074 8 + 0. 006 3 vs 0. 457 6 + 0. 022 6,P^0.05). The CD82 irnmunoreactive cells was 30M in QDDQ- NM1 cell lines and 75~ in QDDQ- NM2 cell lines,the difference between the 2 groups was significant. Conclusions The expression of KAI1/CD82 is negatively correlated with the metastatic potential of human neuroblastoma.
