摘要
目的以自建的人神经母细胞瘤转移瘤细胞（QDDQ-NMl）及原位瘤细胞（ODDQ-NM2）为研究对象探讨KAll／CD82在不同转移潜能人神经母细胞瘤细胞系细胞中的表达情况。方法采用RT-PCR检测两神经母细胞瘤细胞系细胞KAllmRNA的表达；免疫组化方法检测细胞（、D82的表达。结果RT-PCR显示QDDQ-NMl细胞系KAllmRNA的相对含量为0．0748±0．0063，QDDQ-NM2细胞系为0．4576±0．0226，两者比较差异有统计学意义（P〈0．05），免疫组化法检测两细胞系细胞CD82蛋白表达的阳性率分别为30％和75％，两者的差异有统计学意义（P〈0．05）。结论神经母细胞瘤细胞系的细胞高表达KAll／CD82，与人神经母细胞瘤细胞系细胞的体外高转移潜能呈负相关。
Objective To study the expression of KAI1/CD82 in the human neuroblastoma with different metastatic potential. Methods Neuroblastoma NB metastatic tumor （QDDQ-NM） and primary tumor （QDDQ-NM2） cell lines were used in this study. The mRNA expression of KAI1 was determined by reverse transcription-polymerase chain reaction （RT-PCR）. The protein level of CD82 was studied by immunocytoehemical staining. Results The mRNA relative expression level of KAI1 in QDD~ NM1 cell lines was 0. 074 8 -＋ 0. 006 3, which was significantly higher than that in QDDQ- NM2 cell lines （0. 074 8 ＋ 0. 006 3 vs 0. 457 6 ＋ 0. 022 6,P^0.05）. The CD82 irnmunoreactive cells was 30M in QDDQ- NM1 cell lines and 75~ in QDDQ- NM2 cell lines,the difference between the 2 groups was significant. Conclusions The expression of KAI1/CD82 is negatively correlated with the metastatic potential of human neuroblastoma.
