摘要
目的:探讨玻璃体腔内注射外源性CNTF(ciliary neurotrophic factor,睫状神经营养因子),通过检测GAP-43表达观察其对大鼠视神经损伤后视网膜节细胞的保护作用。方法:采用无创血管夹对成年大鼠造成视神经中度不全损伤,实验组伤后即时,1、2、4wk时分别向玻璃体腔内注入人重组睫状神经营养因子溶液2μL(1μg/1μL)。同实验组操作,对照组在每个时间点向玻璃体腔内注人2μL三蒸水,在伤后1,2,4,8wk时分别做生长相关蛋白-43的免疫组织化学检测。结果:伤后1 wk,实验组和对照组在GCL(gallglion cell lgyyy,视网膜神经节细胞层)生长相关蛋白-43的表达呈阳性。实验组与对照组间无统计学意义(P>0.05),伤后2,4,8wk时实验组与对照组在GCL层生长相关蛋白-43阳性表达至高峰,两组间相比较有统计学意义(P<0.05)。
Objective: Explore intravitreal injection the exogenous of CNTF (ciliary neurotrophic factor, ciliary neurotrophic factor) ,by detecting the expression of GAP-43 observed protective effect of retinal ganglion cells after optic nerve injury in rats. Methods:The optic nerve was crushed by a well defined forceps to set up an animal model with incomplete moderate optic nerve injury in of 2p, g CNTF and control group were injected with distilled water and used as control group at immediately followed time 1,2,4,8 week after the injury. Rats were sacrificed at the first, the second ,4th, 8th week after optic nerve injury, the expression of GAP-43 in retina was determined by immunohistochemical techniques at the first, second, 4th, 8th week after optic nerve injury. RespμLt: The expression of GAP-43 in retina following moderate optic nerve injury in CNTF treating groups was higher than that in the control group at the second,4th ,Sthweek after optic nerve injury(P〈0.05).
