Ln5γ2对胃癌细胞迁移、侵袭的影响

摘要 目的探讨Ln5γ2对胃癌SGC-7901细胞迁移、侵袭的影响及可能机制。方法实验分为对照组、Ln5γ2趋化组、上皮生长因子受体(epithelial growth factor receptor,EGFR)干扰组、空载体组。以Ln5γ2诱导趋化SGC-7901,观察穿过铺和未铺Matrigl胶的Transwell小室滤膜的细胞数目。采用ELISA和免疫荧光方法检测细胞缓冲液中MMP-2和MMP-9浓度以及细胞骨架结构改变。结果 Ln5γ2诱导趋化SGC-7901,穿过铺和未铺Matrigl胶的Transwell小室滤膜的细胞数目明显高于对照组(P<0.01),EGFR干扰后数目与对照组无显著差异(P>0.05)。EGFR干扰组细胞MMP-2和MMP-9浓度下降,细胞骨架结构模糊,伪足形成减少或消失。结论 Ln5γ2可以促进胃癌SGC-7901侵袭迁移,可能机制为通过EGFR改变细胞骨架结构,影响MMPs的分泌。 Objective To observe the effects and possible mechanism of Ln5γ2 on the invasion and migration in SGC-7901 cells.Methods Four groups of gastric cancer SGC-7901 cells were studied,including an Ln5γ2 chemotaxis group,an EGFR interference group,an empty vector group and a control group.After Ln5γ2 chemotaxis of SGC-7901 cells,cell numbers passing through the Transwell membrane with or without Matrigel glue were observed.The concentrations of MMP-2 and MMP-9 in the cell buffer were tested by ELISA.The cell skeleton changes of the four groups were examined by laser co-focal microscopy.Results The numbers of cell passing through the Transwell membrane with or without Matrigel glue in the Ln5γ2 chemotaxis group were greater than those of other groups(P0.01).However,the numbers showed no difference after EGFR interference(P0.05).MMP-2 and MMP-9 contents in the EGFR interference group declined obviously.The structure of cell skeleton was faintness and the formation of pseudopodium was reduced.Conclusion Ln5γ2 improves the invasion and migration of SGC-7901 cells.The possible mechanism is Ln5γ2 binding to EGFR,then changing the cell skeleton and reducing MMPs secretion.
