摘要
目的探讨Claudin-23表达与胃癌生物学行为的相关性。方法采用免疫组化SP法检测胃腺癌侵袭转移过程中Clau-din-23的表达。结果胃腺癌细胞表现出不同程度的Claudin-23胞膜染色异位至胞质的迷离表达。黏膜层胃腺癌细胞Clau-din-23迷离表达的程度与患者的性别、年龄、浸润深度及淋巴结转移存在相关性(P<0.001);Claudin-23表达与胃腺癌组织学分化程度相关,分化越高迷离表达程度越低(P<0.001);侵袭前沿区胃腺癌上皮较黏膜层区迷离表达程度加剧,而淋巴结转移灶较侵袭前沿区迷离表达的程度降低(P<0.05);黏膜层区及肿瘤中心胃腺癌细胞Claudin-23重度迷离表达者较轻度迷离表达者术后生存率低(P<0.01)。结论 Claudin-23迷离表达参与胃癌的发生、发展,并影响其预后转归。
Purpose To investigate the relationship of the expression of Claudin-23 with the biological behavior in gastric carcinomas. Methods Immunohistochemistry on tissue microarray was used to detect protein expression of Claudin-23 in gastric carcinomas. Re- suits In contrast to normal epithelium, gastric carcinoma tissues showed different degrees of aberrant expression of Claudin-23 from membrane translocated to cytoplasm. The Claudin-23 aberrant degree in the mucosal layer of gastric carcinoma cells was significantly correlated with age, sex, the invasive depth and status of lymph node metastasis (P 〈 0. 001 ). The expression of Claudin-23 correlated with the differentiated degree of gastric carcinomas, that is, the higher differentiation the lower expression of Claudin-23 ( P 〈 0. 001 ). The Claudin-23 aberrant degree in the invasive front area of gastric adenocarcinoma cell was higher than that in mucosa, but in lymph node metastases lower than that in invasive front area ( P 〈 0. 05 ). Postoperative survival rate in case with Claudin-23 higher aberrant expression in mucosal layer and center area of gastric carcinoma was lower than that in Claudin-23 lower aberrant expression (P 〈 0. 01 ). Conclusions Aberrant Claudin-23 expression is involved in the progression of gastric adenocarcinoma, and affects prognosis of the patients.
