摘要
目的探讨成骨生长肽(OGP)和胰岛素样生长因子1(IGF-1)对体外培养的人牙周膜细胞(hPDLC)碱性磷酸酶活性的影响。方法不同浓度的OGP和IGF-1与体外培养的人牙周膜细胞单独或联合应用,用碱性磷酸酶活性检测法检测细胞内碱性磷酸酶活性。结果OGP可使hPDLC碱性磷酸酶活性明显增强,第3天的最大显效浓度为1×10^-9mol/L,最小显效浓度为1×10^-11mol/L。IGF-1也可增强hPDLC碱性磷酸酶活性,第3天的最大显效浓度为100ng/ml,最小显效浓度为1ng/ml。在第1、3、6天,OGP与IGF1最大显效浓度联合和最小显效浓度联合,对hPDLC碱性磷酸酶活性的增强作用均较对照组增加。结论OGP和IGF.1均可使hPDLC碱性磷酸酶活性提高,在一定范围内呈浓度时间依赖关系,二者联合具有协同效应。
Objective To explore the effect of osteogenic growth peptide (OGP) and insulin-like growth factor-1 (IGF-1) on alkaline phosphatase activity of human periodontal ligament cell (hPDLC) in vitro. Methods Human PDLC were cultured in vitro and were treated with OGP and IGF-1 at different concentrations, and the ALPase activity was measured by enzyme kinetic methods. Results OGP significantly increased the ALPase activity of human PDLC with the largest effectual concentration of 1 ×10^-9mol/L and the minimum concentration of 1 × 10^-11 mol/L at 3 days, IGF-1 promoted the ALPase activity of human PDLC with largest effectual concentration 100 ng/ml and the minimum concentration 1 ng/ml at the same day. Moreover, compared with the control group, OGP combined with IGF-1 at both largest and minimum effectual concentration respectively could more significantly increase the ALPase activity of human PDLC at 1,3 and 6 days. Conclusion Both of OGP and IGF-1 can promote the alkaline phosphatase activity of human PDLC in vitro, showing concentration and time dependence features in a certain range and synergy effect.
