摘要
目的探讨大鼠创伤性脑损伤(TBI)后脑组织中β-微管蛋白(β-tubulin)的表达规律及意义。方法雄性SD大鼠56只,按随机数字表法分为正常对照组(8只)和TBI组(48只);采用头颅侧向旋转致伤方法制作TBI模型,按建模后处死时间分为6 h,12 h,24 h,72 h、7 d,14 d共6个亚组,每组8只动物。采用SP免疫组化方法检测β-tubulin在大鼠脑组织中表达及分布。β-tubulin阳性结果判断标准以胞浆内出现棕黄染色为阳性,反之为阴性,并设阴性对照切片。结果大鼠脑组织中β-tubulin在皮层与海马组织细胞的胞浆、轴突、胞体树突中均有表达,脑损伤后6 h时间点皮层与海马细胞表达均最低(P<0.01),随后表达量逐渐递增,至损伤后14 d其表达量恢复正常值,14 d与正常对照组比较,差异无统计学意义(P>0.05)。结论 TBI后,β-tubulin出现一个表达低值,提示在不同机制下可能对脑损伤的发生与发展发挥重要影响作用。
Objective To investigate the expression and significance of β-tubulin in the brain tissues of rat model of traumatic brain injury (TBI). Methods Fifty-six male SD rats were divided randomly into normal control group (n =8) and TBI group (n =48). The TBI model was established by using lateral head rotation device and subdivided into 6 h, 12 h, 24 h, 72 h, 7 d and 14 d group with 8 in each. The expression and distribution of β- tubulin in the rat brain were investigated immunohistochemically. A negative antibody control had been used in each serial sections to estimate the contribution of non-specific interaction binding. ~sults The expression of β-tubulin was observed in cytochylema, axon and cytodendrite in both of hippocampus and cortex. The expression of β-tubulin decreased to the lowest level in the rat brain tissues at 6 h post TBI (P 〈 0. 01), then increased gradually and returned to normal level at 14 d after TBI. There was no significant difference between TBI group and normal control group in the expression of β-tubulin (P 〉 0. 05). Conclusion The reduced expression of β-tubulin after TBI indicates that β-tubulin may play an important role in the process of brain injury.
