摘要
本文建立了酵母中麦角固醇含量高效液相色谱测定方法。其色谱条件为 ,色谱柱HYPersilBDSC185u反相柱 ,流动相为甲醇 :水 (97∶3) ,紫外检测波长为 2 83nm。酵母样加碱乙醇皂化、提取、洗涤、蒸干、定量测定。结果表明 :标准曲线范围是 0 0 2— 0 8mg/ml线性良好 ,最低限量为0 0 1mg/ml;日内及日间RSD(n =4 )分别在 2 1~ 4 0 %和 2 4~ 4 8% ,回收率为 96 0~ 98 0 %。本方法操作简便、准确、可靠。
A HPLC assay for the determination of ergosterol in yeast was established. A reversed phase C18 column and methanolwater(97∶3) as mobile phase were used,with UV detection at 283nm. The yeast was saponified and then extracted by heptanol.After evaportintg heptanol by vacum the residues were dissolved by ethanol and injected into the system.The results show the calibration curve was linear with the range of 0.02mg/ml~0.8mg/ml.The limit of quantitation was 0.01mg/ml.The average relative deviations (RSD) of within day and between day variation were 2.1%~4.0% and 2.4%~4.8%(n=5) respectively.The recoveries of ergosterol were 96.0%~98.0%.So,this method in a simple,sensitive and accurate method.\;
