实验性变态反应性脑脊髓炎大鼠的中枢和外周儿茶酚胺含量变化 被引量:2

摘要 目的 :探讨实验性变态反应性脑脊髓炎 (EAE)时 ,中枢和外周儿茶酚胺水平的变化规律 ,为神经系统自身免疫性疾病的发病机制和治疗的进一步研究提供实验依据。方法 :用豚鼠脊髓加完全弗氏佐剂制成抗原佐剂乳化物诱导Wistar大鼠发生EAE。在EAE 2级和 3级临床症状时 ,用高效液相色谱 电化学检测法 (HPLC ECD)测定下丘脑、海马、肠系膜淋巴结和血清中去甲肾上腺素 (NA)、多巴胺 (DA)和肾上腺素 (A)的含量。结果 :①诱导大鼠EAE ,一般在注射抗原佐剂乳化物两周后开始出现症状 ,2级症状为后肢无力 ,3级症状为后肢瘫痪 ;②EAE 2级和3级症状时 ,下丘脑和海马中NA含量都明显降低 ,而肠系膜淋巴结中NA含量显著升高 ,且症状越重 ,NA含量变化越大 ;③ 3级症状的EAE大鼠 ,下丘脑、海马和淋巴结中DA含量都显著增加 ,而在 2级症状时 ,只有淋巴结中DA含量明显增加 ;④ 2级症状的EAE大鼠 ,血清中A含量高于正常大鼠近 2 .5倍 ,3级症状的EAE大鼠 ,血清中A含量高于正常近 3倍。EAE大鼠淋巴结中A含量也升高 ,但升高幅度没有血清中的大。结论 :大鼠EAE疾病时 ,主要伴有中枢NA降低 ,外周儿茶酚胺升高 ,且EAE症状越重 ,儿茶酚胺含量的变化越大 ,这可能有利于EAE疾病的恢复。 Aim and Methods:Experimental allergic encephalomyelitis(EAE) was induced with the guinea pig spinal cord in Freund's complete adjuvant,and the changes of noradrenaline (NA),dopamine (DA)and adrenaline (A) were determined in the hypothalamus, hippocampus,lymphaden and serum of Wistar rats at EAE clinical degree two and three in order to demonstrate the relationship between the catecholamines and disease course of EAE.Results:①EAE clinical signs accompanied by demyelin alterations in the central nervous system (CNS) appeared about on the 15th day and the peak signs were on the 19th day after the Wistar rats were injected antigen.The clinical sign of weak hind limbs was marked as EAE symptom degree two and the sign of paralytic hind limbs marked as EAE symptom degree three.② NA content in the hypothalamus and hippocampus significantly decreased and markedly increased in the mesenteric lymph node of EAE rats.The NA changes were stronger at symptom degree three than at symptom degree two.③ Increased DA levels were observed in the hypothalamus,hippocampus and mesenteric lymph node at clinical degree three, whereas at clinical degree two,only in the mesenteric lymph node, DA was significantly elevated.④ In EAE rats,there was a striking augmentation of serum A content which was nearly 2.5 times at clinical degree two and nearly 3 times at clinical degree three as high as that in normal rats.Conclusion:EAE disease is mainly accompanied by NA reduction in CNS and catecholamine enhancement in periphery there are a positive correlation between catecholamines and disease severity,which may contribute to recovery.
