摘要
目的 应用人巨细胞病毒 (HCMV )建立先天性中枢神经系统感染小鼠模型。以电镜研究探讨HCMV先天性感染对中枢神经系统损伤的机制和程度。方法 将 HCMV腹腔内接种纯系 6~ 8周龄Balb/ c雌雄小鼠成功后给予交配受孕 ,待雌鼠临产时取出胎鼠脑双侧大脑皮层 ,进行病毒分离、病理学检验和电镜研究。结果 在鼠脑组织上清液中分离出 HCMV;病理学证实 ,鼠脑为侵袭性脑膜脑炎性病理改变 ;电镜下研究 ,在感染组子代鼠脑组织中找到了人类疱疹病毒样颗粒和神经细胞浆内包涵体。并发现神经元细胞变性、水肿、结构破坏以及脑组织血管内皮细胞凋亡 ;且感染雌鼠所生子鼠死胎率及生后一周内病死率明显高于正常对照组 (P<0 .0 5 )。结论 HCMV能侵袭 Balb/ c小鼠 ,并通过胎盘感染子代的中枢神经系统 (CNS)。这种模拟人类 HCMN先天性 CNS感染小鼠模型的建立所显示许多相似的感染和病理过程 ,为进一步研究 HCMV先天性 CNS感染的病理过程 ,以及应用疫苗来预防孕妇和胎儿先天性感染提供了理论基础 ,对预防神经残疾儿的出生 。
Objective We first established the mouse model that central nervous system was congenitally intected with human cytomegalovirus (HCMV),and then investigated injuried mechanism and degree of central nervous system caused by HCMV.Methods HCMV was injected into intraperitoneum of the mice those were about 6 to 8 weeks old. Then we made the males and the females of the mice copulate and the females be pregnant. When the females were near childbirth,the cerebral cortex of their fetus were removed from the skull and fixed with 4%paraformaldehyde,cut into serial coronal sections 5 um thick,and then stained with hematoxylin and eosin. At last the virus was isolated from brain tissues of the fetus mouse and the cortex of the fetus were examined by labelling and electronic microscopy.Results The viral particles were found in the brain tissues of the fetus, The cortex of the fetus were found out to have large basophilic and intranuclear neuronal inclusions of viral type. Conclusion The research showed that HCMV could not only infected the mice but also infected the fetus.The establishment of mouse model took after the process of infection of buman being.We thought our study was of singnficance for prevention and treatment of HCMV infectien.
