蒜薹成熟期石蜡切片方法研究及解剖学观察 被引量:1

摘要 建立了一种适合大蒜蒜薹成熟期的石蜡制片方法和鉴别品种办法。对取材、固定、脱水、透明、浸蜡、包埋、切片、染色等重要制片环节进行了研究,得到了适合蒜薹成熟期的石蜡制片方法。蒜薹的最佳制片条件为:在白绿相间处取长0.3mm;切片厚度为10μm;1%番红溶液(50%乙醇配制)染色10 min,固绿0.5%(95%乙醇配制)染色2~3 min。镜下可见蒜薹的基本组织分为皮层和髓部两部分。皮层组织由外向里依次是角质层、表皮、分泌组织和周缘维管束;髓部由薄壁细胞和中央维管束组成。该制片方法可得染色清晰、组织结构完整的蒜薹成熟期的石蜡横切片,其显微特征可作为蒜薹的品种鉴定和质量标准制定的参考依据。 In this paper, paraffin sections making suitable for garlic, scape in its maturity and its use for cuhivar identification were established. The key steps such as material selection, fixation, dehydration, transparent, waxing, embedding, section, staining for paraffin section were described. The suitable method for making paraffin sections of garlic scape in its maturity was proposed, and the best conditions for making the paraffin sections of garlic scape were found. Practice proves that a good quality paraffin section with thorough staining, complete structure of cross-section of garlic scape in its maturity stage can be achieved using this method. The microscopic features of the section could provide reference for the cuhivar identification of the garlic scape and the development of its quality standard.
