摘要
目的:探讨三氧化二砷(As2O3)抑制HL-60细胞端粒酶hTERT表达的分子机制。方法:构建含有hTERT启动子不同片段的荧光素酶基因表达载体,转染HL-60细胞,分别在有无As2O3作用下检测各组荧光素酶活性变化,间接反映As2O3对hTERT启动子不同片段活性的影响。结果:在As2O3作用下,含有hTERT启动子不同片段的荧光素酶基因表达载体的荧光素酶活性均下降(P<0.05~P<0.01)。结论:As2O3对HL-60细胞端粒酶hTERT启动子活性具有抑制作用。
Objective:To investigate the molecular mechanism of arsenic trioxide(As2O3) inhibiting telomerase human telomerase reverse transcriptase(hTERT) expression in HL-60 cells.Methods:The different fragments of hTERT promoter luciferase gene expression vector were constructed and transfected into HL-60 cells with or without As2O3.The changes of luciferase activity which indirectly reflected the role of As2O3 on hTERT promoter different fragments were detected.Results:The luciferase activities in different fragments of hTERT promoter gene expression vector were decreased with As2O3(P0.05 to P0.01).Conclusions:As2O3 can inhibit telomerase activity of hTERT promoter in HL-60 cells.
