滋补肝肾方药对局灶性脑梗死大鼠健侧脑组织RhoA、ROCK-2表达的影响 被引量:1

Effect of tonifying the liver and kidney method on expression of RhoA and ROCK-2 in the contralesional cortex of model rats with focal cerebral ischemia

摘要 目的:探讨滋补肝肾方药复健片对局灶性脑梗死大鼠健侧大脑皮质RhoA、ROCK-2基因和蛋白表达的影响。方法:Wistar大鼠随机分为假手术组、模型组和药物组,各组根据观测时间点再分为术后4周末、术后5周末两个亚组。采用电凝法闭塞模型大鼠右侧大脑中动脉,采用RT-PCR方法观测RhoA mRNA、ROCK-2 mRNA表达,采用Western-blot观测RhoA、ROCK-2蛋白表达。结果:在各观测时间点,三组左侧大脑运动皮层组织中RhoA基因和蛋白表达均无显著性差异;而模型组ROCK-2的表达则在术后4周末低于假手术组(蛋白表达,P<0.05;基因表达,P<0.01),至术后5周末,与假手术组已无显著性差异;药物组ROCK-2的表达则在术后4周末和5周末均低于其余两组(与假手术组相比,P<0.01;与模型组相比,除在术后4周末蛋白表达,P<0.01,其余均为P<0.05)。结论:局灶性脑梗死后,未损伤侧大脑皮质的ROCK-2基因和蛋白表达均下调,复健片可抑制相应大脑皮质的ROCK-2表达,这可能是复健片促进未损伤侧大脑重塑的重要机制之一;而RhoA的表达虽然未见明显变化,但不排除活性发生变化。 Objective:To investigate the effect of Fujian tablet tonifying the liver and kidney on the expression of RhoA and ROCK-2 in the contralesional cortex of model rats with focal cerebral ischemia. Methods :Wistar rats were randomly divided in- to three groups:sham surgery group,model group and administration group. Each group were randomly divided into two sub- groups according to observation time including the end of 4 weeks and the end of 5 weeks after operation. The right middle cerebral artery of model rat was occluded by electric coagulation. Gene and protein expression of RhoA and ROCK-2 were de- tected respectively by RT-PCR and Western-blot methods. Results :There were no differences in the expression of gene or pro- tein of RhoA in the contralesional cortex between these three groups. The expression of ROCK-2 in model group was weaker than sham operation group only at the end of 4 weeks after operation(P〈0.05 in protein expression and P〈0.01 in gene expression) and the expression of ROCK-2 in administration group was weaker than other two groups at the end of both 4 weeks and 5 weeks (vs sham group,P〈0.01, vs model group P〈0.05,except at the end of 4 weeks in protein expression,P〈0.01). Conclusion:The expression of ROCK-2 reduced in contralesional cortex of rats with focal cerebral ischemia and Fujian tablet can inhibit the expression of ROCK-2,whieh maybe an important mechanism of Fujian tablet to promote eontralesional cortex plasticity. But it is not proved whether activity of RhoA in eontralesional cortex was changed by cerebral isehemia.
