慢性炎症因子在多囊卵巢综合征发病中的作用 被引量：13

摘要 目的:探讨慢性炎症因子白细胞介素18(IL-18)在多囊卵巢综合征(PCOS)发病过程中的作用。方法:采用不同浓度的IL-18(0、100、200、500、1 000 pg/ml)对体外培养的大鼠颗粒细胞甾体激素分泌、雄激素活性及芳香化酶活性的影响。结果:与对照组(0 pg/ml)相比,IL-18能够抑制颗粒细胞孕酮的分泌(P<0.05),增加Western blot中雄激素受体蛋白的表达,并降低芳香化酶CYP19蛋白的表达;通过qPCR实验发现,在IL-18作用下,颗粒细胞的ARmRNA表达明显增加(P<0.05);CYP19mRNA在500、1 000 pg/ml的IL-18作用下表达呈下降趋势(P<0.05)。结论:IL-18能够抑制颗粒细胞孕酮的分泌,抑制CYP19蛋白及mRNA的表达,抑制芳香化酶活性;并促进AR蛋白及mRNA的表达,间接增加雄激素的活性,可能与慢性炎症参与PCOS的发病有关。
