摘要
目的:探讨抗D抗体的产生与RHD基因Rhesus boxes及外显子的相关性。方法:收集有抗D抗体的血液样本42例,血型血清学方法检测Rh表型,间接抗球蛋白试验确定RhD阴性,吸收放散试验确定RhDel型,聚合酶链式反应-序列特异引物技术检测RHD基因Rhesus boxes及外显子。结果:在42例样本中,24例(57.14%)样本只含有杂交盒,18例(42.86%)样本中同时含有上游盒、下游盒和杂交盒;RHD基因10个外显子全部缺失的有23例(54.76%),部分缺失的有10例(23.81%),所有外显子全部存在的有9例(21.43%)。结论:RhD阴性女性经妊娠产生了抗D抗体,很可能与RHD基因的杂交Rhesus box及RHD基因的不完整性有关。
Objective:To discuss the correlation between the production of anti-D and Rhesus boxes and exons of RHD gene. Method: Samples of 42 RhD negative women with anti-D were collected. Serological method was applied for detection of Rh antigens,indirect antiglobulin test was applied for identification of RhD negative,absorption-elution test was applied to detect RhDel, and polymerase chain reaction-sequence specific primer (PCR-SSP) was applied to detect the Rhesus boxes and exons of RHD gene. Result:Among the 42 samples, 24 (57.14 % ) contained Hybrid box,and 18(42.86%) contained Up box,Down box and Hybrid box. ,23(54.76%) lost all 10 exons of RHD gene,10(23.81%) had a partial deletion of them,and only 9(21.43%) carried 10 exons. Conclusion:The production of anti-D in RhD negative women by pregnancy,may has the relationship with Hybrid box and the now integrality of RHD gene.
