摘要
目的观测2型糖尿病(T2DM)患者尿中足细胞数量的变化,并探讨其相关因素。方法选取124例T2DM患者和25例健康体检者(正常对照组,NC组)。其中T2DM患者分为3组:正常蛋白尿组(NA组),微量白蛋白尿组(MA组),大量蛋白尿组(CP组),应用小鼠抗人肾小球足细胞特异性蛋白Podocalyxin(PCX)单克隆抗体,采用间接免疫荧光技术检测尿中PCX阳性染色细胞,计数尿足细胞数量。ELISA方法检测血清C反应蛋白(CRP)等指标。比较各组的尿中足细胞数量、血CRP水平。结果与糖尿病正常蛋白尿组比较,微量白蛋白尿组、大量蛋白尿组的血CRP、尿足细胞数量均升高(P<0.05)。结论 T2DM患者尿中足细胞数量与尿蛋白相关,并随着血CRP的升高而增加。
Objective To detect the amount of urinary podocytes in type 2 diabetic patients and investigate the relevant factors. Methods 124 patients with T2DM and 25 healthy controls (control group, NC group)were selected. The patients were divided into three groups, normoalbuminuria group (NA group), microalbuminuria group (MA group), macroalbuminuria group (CP group). Indirect immunofluorescene with poly clonal antibody against podocalyx in was used to detect urinary podocytes and the number of podocytes was counted under fluorescence microscope. ELISA was used to detect indicators of serum C-reactive protein (CRP). Compare the number of urinary podocytes of each group, CRP levels. Results CRP and the amounts of urinary podocytes were increased in MA group and CP group contrast with NA group. Conclusion There was significant correlation of the amount of urinary podocytes with urinary protein excretion, and the urinary podocytes number consistents with the changes of inflammatory factors CRP.
