高效液相色谱法测定蜂胶胶囊中吡啶甲酸铬的含量

HPLC Determination of Chromium Picolinate in Propolis Capsules

目的建立保健品蜂胶胶囊中吡啶甲酸铬的含量测定方法的检验操作规程(SOP)。方法色谱柱为phenomenexC18(250mm×4.6mm,5μm),乙腈-磷酸盐缓冲液(磷酸氢二钾4.35g,磷酸二氢钾3.40g,加水1000mL溶解)(10∶90)为流动相,流速:1.0mL/min,检测波长为254nm,柱温:35℃,进样量:10μL。结果吡啶甲酸铬在2.0948～104.7424μg/mL范围内具有良好的线性关系,回归方程:Y=1.307×104x-2.512×103,r=0.9999。平均回收率为98.68%(n=9)。结论采用高效液相色谱法测定蜂胶胶囊中吡啶甲酸铬含量的方法简便、准确可靠,可作为蜂胶胶囊的质量控制方法。

Objective To established a SOP for the determination of chromium picolinate in propolis capsules.Methods The HPLC method was performed with a phenomenex C18（250mm×4.6mm,5μm） column,and the mobile phase was acetonitrile- phosphate buffer （dipotassium dehydrogen phosphate 4.35g,potassium dehydrogen phosphate 3.40g,dissolved by 1000mL water） （10 ： 90）;the detecting wavelength was 254nm;column temperature at 35℃,injection volume 10μ L.Results The calibration curve of chromium picolinate was linear within the range of 2.0948 - 104.7424 μg/ mL,the calibration curve was Y=1.307×10^4x-2.512×10^3,r=0.9999;the average recovery was 98.68%（n=9）.Conelusion The method is simple,with HPLC to graphy method for determination of the content of chromium pieolinate in propolis capsule is accurate and reliable,the method can be used for the quality control method of Prnpolis capsules.
