复方降糖滴丸对实验性糖尿病大鼠血糖、血脂的调节作用及机理研究 被引量:1

摘要 目的:研究复方降糖滴丸对实验性糖尿病大鼠血糖、血脂的调节作用及机理。方法采用静脉注射链脲佐菌素,并给以高脂饲料制备实验性糖尿病大鼠模型,以复方降糖滴丸进行治疗,测定模型大鼠的空腹血糖(FBG)、糖化血红蛋白(HbA1c)、血清中胰岛素(Ins)、甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白(HDL-c)、低密度脂蛋白(LDL-c)、胰岛素样生长因子-1(IGF-1)、肿瘤坏死因子-α(TNF-α)、肝中的超氧化物歧化酶(SOD)、丙二醛(MDA)、脏器系数(肝、肾、胰)和病理学。结果:与模型组相比,复方降糖滴丸能降低模型大鼠FBg,HbA1c、Ins、T g,TC、LDL-C、TNF-α;升高IGF-1;增加肝组织SOD、MDA。与金芪降糖阳性组相比,复方降糖滴丸多个指标效果优于前者特别是HbA1c、T g,LDL-C(P<0.05)。复方降糖滴丸组能显著降低糖尿病大鼠肝(P<0.05 or<0.01)、肾(P<0.01)脏器系数,复方降糖滴丸给药组能有效改善糖尿病大鼠的胰岛细胞质量和数量,缓解高脂饲料喂养引起的脂肪肝和肾损害。结论:复方降糖滴丸能有效降低实验性糖尿病大鼠FB g,HbA1c,其作用机制主要表现为改善大鼠体内糖代谢和促进胰岛素分泌,增加胰岛素生长因子,改善胰岛素抵抗;复方降糖滴丸能降低实验性糖尿病大鼠的血脂,其可能的机理为通过降低糖尿病大鼠体内的TC、TG,低密度脂蛋白。复方降糖滴丸也能促进肝脏脂质过氧化物代谢,此外尚能提高大鼠体内TNF-α。 Objective :To investigate the effection and mechanism of compound hypoglycemic pill in type 2 diabetic rats. Methods:Rat models of type 2 diabetes were made by injecting Steptozotocin(27 mg/kg)intravenously accompanied by feeding high - lipid and high - sugar diet. Then the rats were given compound hypolycemic pill, and fasting blood glu- cose ( FBG), glycosylated hemoglobin 1 c ( HbA1 c ), insulin ( Ins ), triglyceride ( TG), total cholesterol ( TC ), high - density lipo cholesterol( HDL - c) , low - density lipo cholesterol( LDL - c) , insulin - like growth gactors - l ( IGF - 1 ), tumor necrosis factor - α ( TNF - α) , liver super oxide dismutase ( SOD ), malondialdehyde ( MDA ) , organ coefficient (liver, kidney, Pancreas) and pathology were as indicators. Results: Compared to the modle group, it showed that the compound hypolycemic pill can reduce the levels of FBG, HbA1 c, Ins,TG, TC, LDL - C, TNF - α while rise the IGF - 1 level and improve the SOD and MDA in liver. Compared to the postive group, compound hypoglycemic pill also perform- anced better on HBA1 c, TG and LDL - c (P 〈 0.05 ). Compound hypoglycemic pill can obviously lower the liver and kid- ney organ coefficient ( P 〈 0.05 or P 〈 0.01 ) , improve the quality and quantity of Islet cells, and ease the fatty liver and kidney damage due to the high - lipid and high - sugar diet. Conclusion : The experiment results indicate the compound hypoglycemic pill can effectively lower the FBG and HbAI c. The main mechanism was to improve glucose metabolism in rats and the promotion of insulin secretion, to increase insulin growth factors, and to improve insulin resistance. It evi- dently reduced their blood fat levels, it could obviously ameliorate their liver peroxidation by reducing TC, TG and LDL -C. Compound hypoglycemic pill also could promote metabolism of liver lipid peroxides, in addition, it also can improve the level of tumor necrosis factor in diabetes rats.
