摘要
目的:研究红景天苷(SDS)对百草枯(PQ)中毒大鼠急性肺损伤(ALI)的治疗作用及其可能机制。方法:将36只SD大鼠随机分为生理盐水(NS)对照组、PQ模型组和SDS干预组,每组12只。PQ模型组和SDS干预组大鼠腹腔内注射PQ,对照组注射等容积NS,SDS干预组同时给予SDS治疗。24 h后测定肺湿/干重比,进行肺组织HE染色,病理学观察,对肺组织中肿瘤坏死因子-α(TNF-α)含量进行测定。结果:病理学观察表明,PQ模型组肺组织水肿、出血和大量的炎性细胞浸润。SDS干预组的肺组织损伤明显减轻,肺泡腔及支气管腔炎细胞及渗出物与PQ模型组相比明显减少。PQ模型组肺湿/干重比与NS组相比明显增加(P<0.05),肺组织匀浆中TNF-α含量显著增加(P<0.05);而在SDS干预组中,肺湿/干重比、肺组织匀浆中TNF-α含量与PQ模型组相比均明显减少(P<0.05)。结论:SDS对PQ中毒所致ALI具有治疗作用,可能是通过抑制肺组织中炎症因子表达而发挥作用。
Objective:To investigate the mechanism of salidroside(SDS) in the treatment of paraquat -induced acute lung injury(ALI) in rats. Methods:Thirty -six male SD rats were randomly divided into three groups:control group (NS group) , paraquat group ( PQ group), SDS treatment group ( SDS group). The ALI model induced by PQ poisoning was pro- duced hy intraperitoneal injection PQ solution,and the rats in NS group were given normal saline solution with the same volume. One hour after the administration, rats in SDS group were given SDS, and PQ group were given normal saline solu- tion with the same volume. Twenty - four hours later, the wet - to - dry (W/D) weight ratios of the lung and the content of tumor necrosis factor -α( TNF - α) were measured, and the pathology of lung tissue by HE staining was also observed. Results:Histological study showed congestion, edema and sequestration of inflammatory cells in the lung tissues in PQ group,but the lung injury was significantly alleviated in SDS group. Compared to NS group, the content of TNF - α was increased greatly (P 〈 0.05 ) in the lung tissues, and the W/D ratio was significantly increased in PQ group. SDS treat- ment inhibited all these increased parameters ( P 〈 0.05 ). Conclusion : SDS has therapeutic effect on the ALI induced by PQ poisoning and the mechanism may be associated with the reduction of the radical injury.
