摘要
目的:优选皮炎洗剂最佳醇提工艺,建立总蒽醌含量的紫外分光光度和总生物碱含量的高效液相色谱测定方法(HPLC法)。方法:以总蒽醌和总生物碱提取率为考察指标,采用L18(37)正交实验方法优选皮炎洗剂醇提工艺。利用显色反应于505 nm波长处测定总蒽醌类吸收度,采用HPLC法测定总生物碱类化合物含量。结果:最佳醇提工艺:8倍量浓度为70%乙醇提取2次,每次2 h。总蒽醌和总生物碱的吸收度与浓度呈现良好的线性关系。结论:该工艺合理、稳定、可行,紫外分光光度法灵敏、快速、成本较低,HPLC法高效,准确,适用于皮炎洗剂中总蒽醌和总生物碱的含量测定。
Objective:To optimize the ethanol extracting technology for Piyan lotion and establish a ultraviolet spectro- photometric method for the determination of anthraquinones and high - performance liquid chromatography method for the alkaloid in Piyan lotion. Method : The extracting technology of Piyan lotion was optimized by orthogonal experiment with the extraction rate of anthraquinones and alkaloid as the markers. Color reaction occurred sensitively when anthraquinones is in the solution and the absorption value of the colored sample solution was measured at 505nm. HPLC was used to deter- mine the content of the alkaloid. Result : The optimum extracting condition was adding 8 times ethanol and extracting 2.0 h/time for 2 times. The absorptivity of anthraquinones and alkaloid were linearly correlated with concentration, respective- ly. Conclusion:The technology is stable and easy to be operated. The method of direct ultraviolet spectrophotometry and HPLC is accurate, sensitive, quick and cost effective, it can be used to determine total anthraquinones and alkaloid in Piy- an lotion.
