摘要
目的:建立五指毛桃药材中补骨脂素的高效液相色谱测定方法及微量元素的原子吸收光谱测定法。方法:采用反相高效液相色谱法测定补骨脂素的含量,Agi l ent Hypersi C18色谱柱(4 mm×250 mm,5μm),流动相:乙腈-水(35∶65),流速为1.0 mL/mi n,柱温:35℃;检测波长:222 nm。采用火焰原子吸收光谱法测定Ca、Mg、Mn、Fe、Zn、Cu、Pb 7种微量元素的含量,综合比较药材内在质量的优劣。以补骨脂素及各微量元素的含量为指标,应用SPSS软件对不同产地药材进行主成分分析。结果:补骨脂素在0.11~1.19 g范围内,线性关系良好,平均回收率为101.1%,RSD为2.8%。通过对不同产地药材的微量元素的测定,结果显示:质量较优三产地排名为:河源、翁源、清远;质量较差有2个产地:郁南、玉林;其余4个产地质量中等。结论:五指毛桃药材中补骨脂素的HPLC分析方法结果准确,重现性好,可作为五指毛桃药材的质量评价方法。以指标性化学成分及微量元素的含量,结合统计分析方法,能较全面客观地评价五指毛桃药材的质量。
Objective: To establish the determination methods for psoralen of WuZhi MaoToo(Ficus simplicissima Lour.) by HPLC and microelements by atomic absorption spectrometry. Method: Content of psoralen was detected with the method of reversed phase high-performance liquid chromatography (RP-HPLC), Agilent Hypersi C18 column (4 mm× 250 mm, 5 μm) was adopted at the temperature of 35℃. The mobile phase was acetonitrile-water (35:65) at the flow rate 1.0 mL/min. The detection wavelength was 222 nm. The contents of Ca, Mg, Mn, Fe, Zn, Cu and Pb were determined with atomic absorption spectrometry to compare the quality of herbs comprehensively. Principal component analysis of herbs from different producing areas was performed with SPSS software by taking con- tents of psoralen and microelements as indexes. Result: Linearity of psoralen was obtained in the range of 0.11- 1.19 g, average recovery rate was 101.1% and RSD 2.8%. The results of microelements determination in herbs from dif- ferent producing areas revealed that the top three producing areas were Heyuan, Wengyuan and Qingyuan; the two ones with poor quality were Yunan and Yulin; the other four places were with middle quality. Conclusion: HPLC method for psoralen in WuZhiMaoTao was accurate and reproducible, which could be used for quality control of WuZhiMaoTao. Chemical ingredient indexes, the contents of microelements combined with statistical analysis could reflect the quality of WuZhi MaoTao comprehensively and objectively.
