CHIP在人颅内动脉瘤瘤壁的表达及意义

摘要 目的:观察Hsc70羧基末端反应蛋白(CHIP)在人颅内动脉瘤内的表达情况,检测其对瘤壁血管平滑肌细胞凋亡的作用,揭示CHIP与颅内动脉瘤形成的关系。方法:选择2009年6月至2012年12月在包头医学院第一附属医院显微手术切除的颅内动脉瘤标本18例为研究对象,取脑外伤后内减压术切除的正常脑皮层动脉血管5例作为对照,应用免疫印迹的方法检测标本中CHIP和半胱氨酸天冬酶-3(Caspase-3)的表达;通过RT-PCR的方法检测标本内的CHIP mRNA表达。结果:动脉瘤组CHIP的蛋白表达水平低于对照组,Caspase-3的蛋白表达高于对照组;CHIP mRNA在颅内动脉瘤的表达水平低于对照组,差异均有统计学意义。结论:CHIP可能与颅内动脉瘤的形成与生长有关。 Objective: To detect the expression of Carboxyl terminus of Hsc70 - interacting protein (CHIP) on the intracra- nial aneurysm wall and to assess its effect on the apoptosis of vascular smooth muscle cells, aiming at clarifying the relationship between CHIP and the development of intracranial aneurysm. Methods: 18 intracranial aneurysm samples, removed by microsur- gery in the hospital from June 2009 to December 2012, were collected as the experimental group and 5 cases of normal removed pallium artery samples were collected as the control group. Western blotting technique was used to evaluate the expressions of CHIP and caspase - 3. The expression of CHIP mRNA was evaluated by RT - PCR. Results : Compared with the normal group, the expression of CHIP significantly decreased on the intracranial aneurysm walls while the expression of caspase - 3 remarkably increased ( P 〈 0.01 ). Moreover, the expression of CHIP mRNA in intracranial aneurysm samples was lower in the experimen- tal group than that in the control group ( P 〈0.01 ). Conclusion: CHIP may be involved in the formation and the progression of intracranial aneurysm.
