摘要
目的研究穿心莲内酯对人的黑色素瘤细胞(A375)凋亡的影响。方法通过MTT方法检测穿心莲内酯对A375细胞增殖的抑制作用,并求出其半数抑制质量浓度(IC50),并以IC50的穿心莲内酯和衍生物4H-穿心莲内酯作用于A375细胞,并设定溶剂DMSO对照组,Western Blot检测人原癌基因bcl-6的表达,同时用Honechst 33342染液检测A375细胞的凋亡情况。结果穿心莲内酯抑制A375生长的半数抑制质量浓度为30μg/mL;Western Blot检测显示穿心莲内酯明显下调bcl-6的表达;Honechst 33342染色显示穿心莲内酯明显促进了A375细胞的凋亡。结论穿心莲内酯通过下调bcl-6促进A375细胞的凋亡。
AIM To investigate the role of andrographolide on apoptosis in human melanoma cells line A375.METHODS Inhibitory effect of andrographolide on the proliferation of A375 cell line was detected by MTT.IC50 value of andrographolide and 4H-andrographolide was calculated.The solvent DMSO was selected as control group.The expression of bcl-6 was detected by Western Blot and Honechst 33342 dye.RESULTS The IC50 vaule of andrographolide detected by MTT was 30 μg/mL.Andrographolide significantly lowered the expression of bcl-6 by western blot.Honechst 33342 staining showed that andrographolide significantly promoted the apoptosis of A375 cell line.CONCLUSION Andrographolide can promote apoptosis in human melanoma A375 cell line by decreasing bcl-6 level.
