摘要
目的探讨绿原酸对体外过氧化氢诱导大鼠髓核细胞凋亡的影响及机制。方法采用体外培养大鼠髓核细胞,传代后取第3代细胞进行实验。分为4组:正常对照组、绿原酸组(90μmol/L)、过氧化氢组(600μmol/L)、绿原酸+过氧化氢组。通过流式细胞仪检测细胞凋亡情况;RT-PCR检测不同组髓核细胞Bcl-2和Caspase-3基因表达。结果与正常对照组比较,过氧化氢(600μmol/L)能非常明显的诱导髓核细胞凋亡(P<0.05),绿原酸(90μmol/L)能降低过氧化氢诱导的髓核细胞凋亡,表现为细胞凋亡率减少(P<0.05),Bcl-2表达增强,Caspase-3表达减弱。结论绿原酸可抑制过氧化氢诱导的髓核细胞凋亡,其作用机制可能与促进凋亡抑制基因Bcl-2的表达及抑制Caspase-3表达有关。
AIM To explore the influence and mechanism of chlorogenic acid on H2O2 induced apoptosis of rat nucleus pulposus cells(NPCs) in vitro.METHODS The passage 3 NPCs that were isolated from rat were divided into four groups,normal control group,chlorogenic acid group(90 μmol/L),H2O2 induced apoptosis group and chlorogenic acid protected group.Flow cytometer test was used to observe rat NPCs apoptosis,and RT-PCR was used to detect the expressions of Bcl-2 gene and Caspase-3 gene.RESULTS Compared with normal control group,H2O2(600 μmol/L) could obviously induce apoptosis in NPCs(P0.05).Chlorogenic acid(90 μmol/L) could decrease the percentage of H2O2 induced NPCs apoptosis(P0.05).Expression of Bcl-2 was enhanced,and expression of Caspase-3 reduced.CONCLUSION The experiment shows that chlorogenic acid can inhibit H2O2 induced apoptosis of rat NPCs,and its mechanism may be related to promoting the apoptosis inhibiting gene Bcl-2 and inhibiting Caspase-3 expressions.
