滋肾养阴汤对去卵巢大鼠性激素水平及抗氧化能力的影响

Effects of Zishen Yangyin Decoction on sex hormones levels and antioxidant capacity of ovariectomized rats

目的研究滋肾养阴汤(生地、山茱萸、淮山药、丹皮、泽泻、黄精、菟丝子、茯苓、女贞子、旱莲草)对去卵巢大鼠性激素水平和抗氧化能力的影响。方法除对照组外,切除成年雌性大鼠双侧卵巢,将模型大鼠随机分为模型组、阳性对照组和滋肾养阴汤高、中、低剂量给药组。采用放免法(RIA)测定给药四周后大鼠血清雌二醇、促黄体生成素、促卵泡生成素和孕酮水平的变化。同时检测大鼠体质量及血清中丙二醛(MDA)、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)水平。结果滋肾养阴汤可以显著减少去卵巢大鼠的体质量增加;显著升高血清中雌二醇、孕酮水平,降低促卵泡生成素、促黄体生成素水平;还可以显著升高血清中SOD、GSH-Px水平,降低MDA水平。与模型组比较均有显著性差异(P<0.05或P<0.01)。结论滋肾养阴汤可抑制去卵巢大鼠肥胖,对体内雌激素水平有一定调节作用,并提高其抗氧化能力,可以用于改善女性围绝经期症状。

AIM To study the effets of Zishen Yangyin Decoction（Rehmanniae Radix,Corni Fructus,Dioscoreae Rhizoma,Moutan Cortex,Alismatis Rhizoma,Polygonati Rhizoma,Cuscutae Semen,Poria,Ligustri lucidi Fructus and Ecliptae Herba） on sex hormones levels and antioxidant capacity of ovariectomized rats.METHODS Bilateral ovariectomy was performed in the adult female rats except the sham operation group,and the model rats were randomly divided into model group,positive control group,and groups of high-dose,medium-dose and low-dose Zishen Yangyin Decoction.The changes in the serum levels of estradiol,prolan B,follitropin alfa and progesterone in rats were determined after four weeks by RIA.At the same time,the body weight and the contents of malonaldehyde（MDA）,superoxide dismutase（SOD） and glutathione peroxidase（GSH-Px） were measured.RESULTS Zishen Yangyin Decoction was capable of restraining significantly the weight increase of the ovariectomized rats,significantly raising the levels of estradiol and progesterone in the serum,decreasing the levels of follitropin alfa and prolan B,significantly increasing the levels of SOD and GSH-Px in serum,and decreasing the level of MDA.There were significant differences among the model group and other groups（P0.05 or P0.01）.CONCLUSION Zishen Yangyin Decoction is conducive to the inhibition of the obesity among the ovariectomized rats,up-regulatory effects on the level of female hormones and antioxidant capacity,and the improvement in the perimenopausal syndrome.
