摘要
目的探索薏苡仁提取物对酪氨酸酶抑制作用。方法采用多巴色素法测定薏苡仁提取液对酪氨酸酶的抑制作用,正交设计优化薏苡仁提取物的提取工艺;采用酶促动力学方法探讨薏苡仁提取物对酪氨酸酶的抑制作用类型。结果加8倍量的70%乙醇提取2 h,提取两次得到的薏苡仁提取物对酪氨酸酶的抑制率达33.3%。薏苡仁提取液的半抑制质量浓度(IC50)为1.4 mg/mL。Lineweaver—Burk双倒数作图显示,薏苡仁提取物对酪氨酸酶的抑制作用属于线性混合型抑制。结论薏苡仁提取液具一定的美白作用。
AIM To explore the inhibition of Coicis Semen extract on tyrosinase.METHODS The inhibitory effect of Coicis Semen extract on tyrosinase activity was determined by Doba pigment method.The orthogonal test design was used to optimize the extraction conditions of enzyme-catalyzed reactions to study the type of inhibition kinetics.RESULTS The optimum extraction process was 70% ethanol,weightvolume of 18,2 hour of extraction time,twice,and the inhibitory rate was 33.3%.The concentration of Coicis Semen extraction was 1.4 mg/mL leading to 50% activity lost(IC50).The Lineweaver-Burk plot indicated that Coicis Semen extract was the linear mixed-type inhibitor.CONCLUSION Coicis Semen extract has a role in whitening effect.
