骨髓干细胞分化的成骨样细胞与可降解镁合金体外相容性的研究

摘要 目的探讨SD大鼠骨髓间充质干细胞诱导分化的成骨样细胞在可降解镁合金支架材料表面的生长情况,为研究镁合金与成骨样细胞的体外相容性提供实验数据。方法原代培养大鼠骨髓干细胞,并诱导其分化为成骨样细胞,行碱性磷酸酶(ALP)染色鉴定;将成骨样细胞支架材料复合培养,扫描电镜观察细胞形态;观察材料浸提液培养下细胞的形态,及增殖情况,ALP活度检测细胞的成骨分化情况。结果骨髓干细胞呈长梭形,可分化为成骨样细胞,碱性磷酸酶染色阳性。成骨样细胞与镁合金材料复合培养后,扫描电镜显示细胞生长旺盛,镁合金材料浸提液可促进大鼠成骨样细胞生长、增殖和分化。结论可降解植骨材料镁合金与骨髓干细胞来源的成骨样细胞具有良好的组织相容性,有利于细胞黏附、生长增殖和成骨分化。
