摘要 探索微水环境中酸性蛋白酶催化蔗糖和乙酸乙烯酯的酯化反应,对催化合成蔗糖乙酸酯的反应溶剂进行比较,确定四氢呋喃为较好的反应介质,并详细考察了该反应的参数:缓冲液pH值、含水体积分数、反应温度以及反应时间对酯化效果的影响.确定了该酯化反应的最优工艺条件:缓冲液pH为5.4,反应温度为40℃,反应14h后,每0.68g蔗糖可制得蔗糖乙酸酯30.46mg,蔗糖-6-乙酸酯占总酯质量的61.58%. In order to explore the esterification between vinylacetate and sucrose catalysed by acid protease in trace water system, in this resarch, various reaction solvents were screened, and finally the Tetrahdrofuran was selected as the best solvent. The effects of the reaction parameters were investigated and the optimum operation conditions are obtained as follows- buffer pH=5.4, V (water) : V (Tetrahdrofuran) = 30%, reaction temperature of 40 ℃. Under the optimum conditions mentioned above, every 0.68 g sucrose can introduce 30.46 mg sucrose acetate after 14 h, the selectivity of 6-sucrose acetate reached 61.58%.
