胶原诱导型大鼠类风湿关节炎模型的制备

Preparation of collagen-induced rat rheumatoid arthritis model

目的探讨改良胶原诱导性关节炎(collagen induced arthritis,CIA)模型的制备及鉴定方法;方法将Ⅱ型胶原溶液与等体积完全弗氏佐剂在冰浴状态充分乳化制成Ⅱ型胶原乳剂,将Ⅱ型胶原乳剂注射到大鼠的足跖部皮下,首次免疫一侧一次注入Ⅱ型胶原乳剂0.2 ml,7天后用同样方法加强免疫。造模第21天对观察大鼠的关节炎表现进行关节炎指数评分及组织病理学检查。结果造模组大鼠足跖部出现肿胀,关节明显红肿增粗,活动受限。造模组与正常组关节炎评分比较,P<0.05,两组有显著差异。HE染色,造模组滑膜细胞水肿,有3～5层明显增生,细胞排列不整齐,滑膜组织中有大量的炎性细胞浸润。结论探讨改良的CIA模型能反映类风湿关节炎的临床表现,可以为临床治疗做出理论依据。

Objective：Probe into the impovement in the preparation and identification about the model of collagen induced arthritis（CIA）.Methods：We mingle the elementary typeⅡcollagen solution with Freund adjuvant at the same proportion volume.Tight emulsification during ice-bath.Inject rats on one side of the voix pedis by using typeⅡcollagen emulsification 0.2ml at the first immuniztion,enhanced-immunization with the same measure seven days later.Results：The con-diton of rats model at the 21th-day after rats modle successful induced,analyze arthritis index and histopathology status,Conclusion：We found the voix pedis in the group which was successfully induced was swelling,espesially jionts,had a limitation of motivation.Compared with the vacuity contrast group,two groups were obviously distinguished.HE staining：synoviocytes in model group was dropsical,3-5 layers obvious proliferated,arranged irregularly.Inflammatory cells diffussed widely in the synovium.Probe into the improved CIA modle can relacts clinical feature on arthritis.These model can be used as theoretical basisfor design and practice of clinical treatments.
