摘要
目的研究霉酚酸酯对糖尿病大鼠的保护作用,并探讨其作用机制,对MCP-1、MIF表达的影响。方法选取雄性Wistar大鼠55只,行右肾切除术后两周,随机分成模型组和正常对照组(A组),将糖尿病模型组大鼠随机分为糖尿病大鼠对照组(B组)和糖尿病大鼠治疗组(C组)。应用酶免疫分析法测定大鼠24h尿白蛋白排泄率,采用HE、PAS染色,行肾脏组织形态学分析,用Real time FQ-PCR检测MCP-1、MIF等细胞因子的mRNA表达情况。结果和B组(13.71±0.62)相比,C组大鼠尿白蛋白排泄率明显下降(4.32±0.30),差异具有统计学意义(P<0.05);C组KW/BW、MGV、FMA比B组有不同程度的下降(P<0.05);C组大鼠肾脏MCP-1、MIF的mRNA表达有所下调(P<0.05),但仍高于A组;MCP-1、MIFmRNA表达水平与UAER成正相关(r=0.703,P<0.05;r=0.657,P<0.05);MCP-1mRNA表达水平与KW/BW、MGV、FMA成正相关(r=0.651,P<0.05;r=0.639,P<0.05;r=0.771,P<0.05);MIFmRNA表达水平与KW/BW、MGV、FMA成正相关(r=0.567,P<0.05;r=0.593,P<0.05;r=0.876,P<0.05)。结论霉酚酸酯对糖尿病大鼠肾脏可起到保护作用,其机制可能与下调MCP1、MIFmRNA的表达有关。
Objective:To assess the renal protective effects and the expression of MCP1mRNA and MIF mRNA of myclate mofetil(MMF) in diabetic model rats.Methods:55 Male Wistar rats were randomized into the following groups:(A) normal control;(B) diabetic control;(C)diabetic treatment group(MMF group).24h urinary albumin excretion rate(UAER) was detetcted by Enzyme immunoassay;The renal patho-logical changes of renal tissue were observed by HE,PAS stain;Expressions of MCP-1 and TIMP-1mRNA in renal tissue were detected by Real time FQ-PCR.Results:UAER of group C(4.32±0.30)was significantly lower than that of group B(13.71±0.62)(P 0.05);KW/BW,MGV and FMA of group C were significantly lower than that of group B(P 0.05);Expressions of MCP-1 and MIFmRNA of group C were significantly lower than group B,but still higher than group A;and which were possotively correlated with 24h UAER,KW/BW,MGV and FMA(P 0.05).Conclusion:MMF has protective effect to diabetic kidney.The mechanism may be correlated with down-regulating the expression of MCP-1 and MIF.
