摘要
目的探讨Caveolin-1、P63及CK34βE12蛋白在前列腺病变中表达及其临床意义。方法应用免疫组化染色采用链霉菌抗生物素蛋白-过氧化物酶连结法(streptavidin-perosidase,SP)检测59例前列腺癌(prostate carcinoma,PCa)、15例高级别前列腺上皮内瘤变(high prostate intraepithelial neoplasma,HGPIN)、11例低级别前列腺上皮内瘤变(low prostate intraepithelial neoplasma,LGPIN)、7例非典型性腺瘤样增生(atypical adenomamus hyperplasia,AAH)、10例基底细胞增生(basal cell hyperplasia,BCH)、69例前列腺良性增生(benign prostate hyperplasia,BPH)标本中的Caveolin-1和P63及CK34βE12的表达情况。结果 Caveolin-1、P63、CK34βE12抗原在PCa和BPH标本中的表达差异均有统计学意义(P<0.05)。PCa、HGPIN、LGPIN、AAH、BCH、BPH中Caveolin-1阳性表达率依次为:69.49%、40.00%、45.45%、28.57%、30.00%、8.70%;P63阳性表达率依次为8.47%、86.67%、90.91%、85.71%、90.00%、97.10%;CK34βE12为8.47%、93.33%、90.91%、100%、90.00%、95.65%。Caveolin-1表达强弱与PCa的Gleason评分、临床分期相关(P<0.05)。结论Caveolin-1与PCa的发生、发展、侵袭、转移有关,联合检测Caveolin-1、P63及CK34βE12的表达将对前列腺癌的早期诊断准确性、判断病情以及预后有一定临床指导意义。
Objective To investigate the expression and clinical significance of Caveolin-1,P63 and CK34βE12 protein expression in prostate lesions.Methods Immunohistochemical staining for Caveolin-1 P63 and CK34βE12 was performed by using En Vision in 59 cases of prostate carcinoma(PCa),15 cases with high grade prostatic intraepithelial neoplasia(HGPIN),11 cases with low grade prostatic intraepithelial neoplasia(LGPIN),7 cases with atypical adenomatous hyperplasia(AAH),10 cases with basal cell hyperplasia(BCH) and 69 cases with benign prostate hyperplasia(BPH).Results The expression of Caveolin-1,P63,CK34βE12 antigen in prostate cancer and BPH specimens were significantly differentially(P0.05).The positive expression rate of Caveolin-1 in prostate cancer,HGPIN,LGPIN,AAH,BCH and BPH was 69.49%(41/59),40.00%(6/15),45.45%(5/11),28.57%(2/7),30.00%(3/10),8.70%(6/69) respectively,the positive expression rate of P63 was 8.47%(5/59),86.67%(13/15),90.91%(10/11),85.71%(6/7),90.00%(9/10),97.10%(67/69) respectively,and the positive expression rate of CK34βE12 was 8.47%(5/59),93.33%(14/15),90.91%(10/11),100%(7/7),90.00%(9/10),95.65%(66/69) respectively.The expression degree of Caveolin-1 was related with the prostate cancer Gleason score and clinical stage(P0.05).Conclusion The expression of Caveolin-1 is related with the occurrence,development,invasion and metastasis in prostate cancer.Combined detection of the expression of Caveolin-1,P63,and CK34βE12 can be helpful for the early accurate diagnoses and the malignancy and prognosis prediction of the prostate cancer.
