RP-HPLC测定委陵菜中槲皮素含量研究

Study on RP-HPLC Determination of Quercetin in Potentilla chinensis Ser.

[目的]建立测定委陵菜中槲皮素含量的方法。[方法]采用Zorbax SB C18柱(250mm×4.6 mm,5μm),流动相为乙腈-水-磷酸(50∶50∶0.04),检测波长为360 nm,测定委陵菜中槲皮素含量。[结果]槲皮素在0.2～2.1μg/mL(r=0.999 7)范围内与峰面积呈良好的线性关系(n=6);样品平均回收率(n=9)为99.4%,RSD为2.57%。[结论]建立的方法简便、快速、准确,重现性好,可用于委陵菜中槲皮素含量的测定。

[Objeetive] The aim was to establish a method for the deternination of quereetin n Potentilla chinensis Se..[Method] The contents of quercetin in Potentilla chinensis Ser. were determined, with the reversed phase HPLC system consisting of C18 column （250mm×4.6mm,5μm）and a mixture of acetonitrile-water-phosphoric（50：50：0.04）as the mobile phase , and the absorbance was monitored at 360 nm.[Result]The quereetin response range was 0.2-2.1μg/mL （r=0.9997）（n=6）.The average recovery （n=9） of quercetin was 99.4%,and RSD was 2.57%.[Conclusion] The assay demonstrated that the method had adequate accuracy and selectivity to measure the concentrations of quercetin in Potentilla chinensis Ser..
