磷脂酰肌醇蛋白聚糖-5基因在肺鳞癌中的表达及临床意义 被引量:1

Expression and clinical significance of glypican-5 in lung squamous cell cacinomas

摘要 目的探讨磷脂酰肌醇蛋白聚糖-5(GPC-5)在肺鳞癌中的表达变化及其临床意义。方法应用实时定量聚合酶链反应(Real-time PCR)的方法检测64例肺鳞癌组织及其对应的肺正常组织中的GPC-5基因的表达。应用免疫组织化学方法验证GPC-5在肺鳞癌组织芯片(包含75例鱗癌组织标本)中的表达。结果GPC-5 mRNA和蛋白表达水平在肺鳞癌组织中均较正常组织显著下降(P 〈0.01);有淋巴结转移组较无淋巴结转移组GPC-5 mRNA和蛋白表达水平显著降低(29. 0074 ±6. 4013 比 11. 4849 ±2. 5066,p 〈0. 05);病理分期 II A -B 组的 GPC-5 mRNA 表达水平较I A -B 组表达显著下降(28. 0566 ±3. 8333 比 11. 9384 ±6. 6651,P 〈0. 05) ;Spearman 相关分析显示GPC-5 mRNA表达水平与淋巴结转移呈显著正相关(r = 0.270,p〈0. 05)。结论GPC-5可能是1种新的抑癌基因,且与肺鳞癌的淋巴结转移密切相关。 Objective To investigate glypican-5 (GPC-5) expression in lung squamous cell carci-nomas and its clinical significance. Methods GPC-5 was detected in 64 cases of lung squamous cell carci-noma and corresponding normal lung tissue by real-time quantitative polymerase chain reaction ( PCR),which was also validated in human lung squamous cell carcinoma tissue microarrays (TMA, containing 75cases of squamous cell carcinoma tissue and adjacent tissue) by using immunohistochemistry. Results Theexpression levels of GPC-5 mRNA and protein in squamous cell carcinomas were significantly lower than inmatched normal lung tissues (p 〈0. 001 ) . The expression levels of GPC-5 mRNA and protein in the lymphnode metastasis group were lower than those in the non-metastasis group (29. 0074 ±6. 4013 vs 11. 4849 ±2.5066 folder, P 〈0. 05). The expression levels of GPC-5 mRNA in the pTNM II A-B group were lowerthan those in the pTNM I A-B group ( 28. 0566 ± 3.8333 vs 11.9384 ± 6.6651 folder, P 〈 0. 05 ).Spearman analysis revealed that the expression levels of GPC-5 mRNA were significantly correlated withlymph node metastasis ( r = 0. 270 ,P 〈 0. 05 ) . Conclusion GPC-5 may be a new tumor suppressor gene,and is closely related with the lymph node metastasis of lung squamous cell carcinoma.
