全反式视黄酸抑制人大肠癌细胞Ⅳ型胶原酶的分泌

摘要 Ⅳ型胶原酶包括72kDa Ⅳ型胶原酶/MMP-2和92kDa Ⅳ型胶原酶/MMP-9两种类型,主要降解细胞外基质中的Ⅳ型胶原,它在肿瘤细胞中的表达水平与许多恶性肿瘤细胞的侵袭和转移能力密切相关。本研究的目的旨在探讨全反式视黄酸(all-trans retinoic acid,RA)对肿瘤细胞分泌Ⅳ型胶原酶的作用。 TypeⅣ collagenase,consisting of 72kDa and 92kDa type Ⅳ collagenase,have close relationship with tumor invasion and metastasis. Human colorectal carcinoma cells cloneA was treated with all-trans-Retinoic Acid. Gelatin zymog-raphy showed that the activity of both active and latent forms of type Ⅳ collagenases decreased. RT-PCR showed that 92kDa type Ⅳ collagenase gene expression was down regulated. So, we conclude that the secretion of type W collagenases in human colorectal carcinoma cells can be inhibited by RA and the effect probably happens in the level of transcription.
