少动鞘脂单胞菌S1胞外多糖发酵工艺条件研究 被引量:13

摘要 研究了摇瓶培养条件下少动鞘脂单胞菌引的胞外多糖的发酵工艺。S1菌的发酵产胶可采用二步发酵法:第一阶段,培养基成分为:蔗糖10g,NH_4NO_3 0.5g,KH_2OP_4 0.5g,MnSO_4 0.3g,MgSO_4 0.1g,吐温80 0.02g溶于蒸馏水并定容至1000mL,pH7.2±0.1,温度33℃~35℃,高溶氧。第二阶段,发酵 10~12h后,补加蔗糖40g/L,温度 24℃~25℃,pH6.5±0,1,高溶氧(激烈振荡)。S1菌胞外多糖的产量最高可达8.98g/L,发酵液粘度最高可达45000mPa·s。 Nutritional requirements and optimal condition for exopolysaccharide (EPS) produchon by Sphingomonas paucimobilis S1 were studied. A two-stage fermentation process was conducted as follows: in the earlier stage which lasted for 10-12h, the initial medium contained the following: sucrose 10g, NH_4NO_3,0.5g, KH_2PO_4 0.5g, MnSO_4 0.3g, MgSO_4 0.1g, Tween 80 0.02g, which is dissolved in distilled water to 1000ml, the pH and temperature were 7.2 ± 0.1 and 33-35℃ respectively; in the successive stage, 40g/L of sucrose was added into the broth, and the pH and tempethere were 6.5±0.1 and 24-25℃ respectively.In both stages, highly resolved O_2 (vigorous agitation) was required. Maximum yield of the EPS (8.98g/L) and maximum viscosity of the broth (45000mPa· s) were observed under optimal condition.
