摘要
目的:分析ST3Gal唾液酸糖基转移酶家族在不同肝细胞系表达的差异,为ST3Gal唾液酸糖基转移酶家族与细胞膜唾液酸糖链表达相关性研究奠定基础。方法:采用实时荧光定量PCR技术检测人正常肝细胞系L-02、人肝癌细胞系HepG-2、SMMC-7721中ST3Gal唾液酸转移酶家族全部成员ST3GalⅠ~Ⅵ的mRNA表达情况。结果:与L-02相比,HepG-2、SMMC-7721中ST3GalⅣ高表达;ST3GalⅠ、ST3GalⅤ、ST3GalⅥ低表达;ST3GalⅡ、ST3GalⅢ不稳定表达。结论:高表达的ST3GalⅣ可能是调节细胞膜表面唾液酸糖链的关键酶之一,并与肝癌细胞膜表面α23-唾液酸糖链表达增高相关。
Objective: To analysis the differences of ST3Gal sialic acid glycosyltransferase expression between different hepatic cell lines, which was the foundation of the expression correlation study between sialic acid glycosyhransferases and cell membrane carbohydrate chains. Method: All members of ST3Gal sialic acid glycosyltransferaes family ST3Gal Ⅰ-Ⅵ expression on mRNA levels of L-02, HepG-2, SMMC-7721 cell lines were detected by Realtime PCR method. Result: Comparing with the hepatic cell line L- 02, ST3Gal Ⅳ highly expressed in the hepatoma cell lines HepG-2 and SMMC-7721, ST3Gal Ⅰ , ST3Gal Ⅴ, ST3GalⅥ lowly expressed, ST3Gal Ⅱ , ST3Gal Ⅲ were unstably expressed. Conclusion: Overexpresed ST3Gal IV may be one of the key enzyme to regulate the cell surface sialic acids, which participates in the increase of hepatoma cell membrane α2, 3-sialic acids.
