胰腺癌组织RUNX3基因启动子区CpG岛甲基化研究 被引量:1

摘要 目的:检测胰腺癌组织抑癌基因RUNT相关转录因子3(human runt-related transcription factor 3,RUNX3)基因启动子区CpG岛甲基化的状态并探讨其临床意义。方法:采用甲基化特异性聚合酶链式反应(MSP)检测52例胰腺癌组织及癌旁组织、15例正常胰腺组织中RUNX3基因启动子区CpG岛甲基化状态,并分析其与胰腺癌临床病理特征之间的关系。结果:胰腺癌、癌旁胰腺组织及正常胰腺组织中RUNX3基因启动子区CpG岛甲基化率分别为48.1%(25/52)、9.6%(5/52)和0.0%,3组间比较差异有统计学意义(P<0.05);RUNX3基因启动子区CpG岛甲基化状态与胰腺癌的TNM分期、分化程度及淋巴结转移关系密切(P<0.05)。结论:RUNX3基因启动子区CpG岛甲基化是胰腺癌组织区别于正常胰腺组织的分子事件之一,可能在胰腺癌的发生、发展中起重要作用;检测RUNX3基因启动子区CpG岛甲基化可能会成为一种新的诊断胰腺癌的肿瘤标志物。
