摘要
目的 :探讨缺氧对大鼠肺动脉平滑肌细胞的作用机制。方法 :应用免疫组化、流式细胞仪和同位素测定方法研究了缺氧条件下培养的大鼠肺动脉平滑肌细胞(PSMC)的生长特性 ,测定了缺氧时其游离钙浓度 [Ca2 + ]和环磷酸腺苷 (cAMP)、环磷酸鸟苷 (cGMP)含量变化。结果 :①缺氧条件PSMC生长特性有改变 ;② [Ca2 + ]明显升高 ,随时间可持续升高 ;③cAMP呈现先降后升 ,cGMP整体水平上升。结论 :缺氧可能通过第二信使传递系统发挥作用 ,损害PSMC。
To investigate the effect of hypoxia on rat pulmonary smooth muscle cell(PSMC).Methods:Immunocytochemical and cell cycle analysis methods were investigated the changes of PSMC.Results:①The growth circumstance and protein contents of PSMC in hypoxia condition was similar to that in normoxia condition.②[Ca 2+ ] lervel was significantly increased and the inerease was more significant when it was prolonged.③cAMP and cGMP of PSMC was significanitly increased.Conclusions: It suffest that hypoxia exerts influence directly on the second messenger system of PSMC.
