阿尔茨海默病APPswe/PS1dE9双转基因小鼠的行为学研究

摘要 目的:研究APPswe/PS1dE9双转基因小鼠的感觉运动活性/移动探索活性、情感和物体识别能力。方法:采用旷场实验和新颖物体辨别实验分别对6月龄(雌、雄)和12月龄(雄)的APPswe/PS1dE9转基因小鼠进行行为学研究,以同窝同龄同性别野生型小鼠为对照。结果:在旷场实验中,与野生型小鼠相比,双转基因小鼠在12月龄时出现探索活性降低且可能更具有焦虑倾向(P<0.05);而在物体识别任务中,各研究组间差异无统计学意义。结论:APPswe/PS1dE9双转基因小鼠能较好地模拟AD患者的部分精神行为症状,为AD的发病机制和治疗研究提供了有价值的工具。 Objective: To investigate the sensorimotor activity/locomotor activity, emotion disorders and the object-recognition capability of APPswe/PS1dE9 double transgenic (Tg) mice. Methods: Open field test (OFT) and novel object recognition test (NORT) were performed in APPswe/PS 1 dE9 double transgenic mice and their age- and gender-matched wild type (wt) littermates at 6 months (female, male) and 12 months (male) of age re- spectively. Results: Compared to their controls, only 12 month-old Tg mice showed hypoactivity and increased anxiety profile in the OFT (P〈0.05). No significant differences between the Tg and wt mice had been detected in the NORT. Conclusion: APPswe/PS1dE9 double Tg mice can preferably recapitulate some behavioral and psychological phenotypes of AD patients, and hence provide a valuable tool to explore the pathogenesis and effective therapeutic strategies of AD.
