摘要
目的:探讨CD105和血管生成素2(Angiopoietin-2,Ang-2)在大肠癌中的表达和相互关系,以及它们在血管生成中的作用;探讨CD105作为肿瘤新生血管标记物的可能性及临床价值。方法:运用免疫组织化学技术检测CD105,Ang-2和CD34蛋白在80例大肠腺癌组织和40例正常肠组织中的表达情况,并计算微血管密度(Microvessel density,MVD),分析其表达与临床病理因素之间的关系。结果:(1)MVD及Ang-2在大肠癌组织与正常肠组织间表达的差异具有非常显著性意义(P<0.01,P<0.01);(2)以CD105和CD34测得的MVD差异具有非常显著性意义(P<0.01);(3)Ang-2在大肠癌中的表达与CD105的表达呈显著正相关(P<0.01),而与CD34的表达无关(P>0.05);(4)大肠癌组织中以CD105测得的MVD和Ang-2的表达与浸润深度、淋巴结转移及Dukes分期有关,差异具有非常显著性意义(P<0.01);而与性别、年龄及分化程度无关(P>0.05)。结论:(1)CD105和Ang-2均与大肠癌血管生成有关,二者协同在大肠癌生长、浸润及转移过程中发挥重要作用;(2)CD105在大肠癌组织新生血管中有良好表达,以CD105测得的MVD可以被认为是与肿瘤生长、转移及预后有关的一项重要指标;CD105作为大肠癌新生血管有价值的标志物,对于大肠癌的早期诊断、判断预后和综合治疗具有重要临床意义。
Objective: To determine the expression of CD105 and Ang-2 and the correlation between them in colorectal cancer( CRC ), and to determine their effect in angiogenesis. To determine the probability and clinical value of CD105 as a marker of tumor-related angiogenesis. Methods: Immuno-histochemical technique was used to detect the expression of CD105, Ang-2 and CD34 protein in 80 cases of colorectal cancer and 40 cases of normal eolorectal tissues, then to count the microvessel density(MVD) and analysis the relationship between the expssion and clinicopathological parameters. Results: ( 1 ) The MVD and the expression of Ang-2 in CRC tissues were significantly higher than those in normal colorectal tissues(P〈0.01 ). (2) The MVD marked by anti-CD105 in CRC and normal colorectal tissues was significantly higher than that marked by anti-CD34 (P〈0.01). (3) Ang- 2 expression was significantly correlated with CD105 expression, but not with CD34 expression(P〈 0.01,P〉0.05). (4)The MVD marked by anti-CD105 and the expression of Ang-2 were closely associated with invasive depth,lymphonode metastasis and Dukes stages of CRC ( P〈0.01 ), and were not associated with gender ,age and differential degree of CRC(P〉0.05 ). Conclusion:( 1 )CD105 and Ang-2 were associated with angiogenesis of CRC, they had a coordinated effect in the growth, infiltration and metastasis of CRC. (2)The MVD marked by CD105 can be considered as an important index which is associated with tumor growth, metastasis and prognosis. CD105 used as an valuable marker of angiogenesis has important clinical significance for early diagnosis, judgement prognosis and complex treatment of CRC.
