摘要
目的 研究 VP- 1 6诱导 PC- 3细胞凋亡和癌基因 bcl- 2及 C- myc表达的关系。方法 用末端标记法(TUNEL )和流式细胞术 (FCM)研究 PC- 3细胞凋亡 ,以免疫组化方法研究 bcl- 2及 C- myc蛋白的表达。结果 TUNEL和FCM检测表明 ,PC- 3细胞的凋亡率随 VP- 1 6作用浓度增加和作用时间延长而升高 ;免疫组化结果显示 :bcl- 2和 c- myc基因表达的阳性百分率随 VP- 1 6作用浓度增加和作用时间延长而降低。结论 VP- 1 6能够诱导 PC- 3细胞凋亡和抑制其bcl- 2和 c- myc基因的表达 ,且具有作用浓度和作用时间依赖性。
Objetive To study the relationship between apoptosis induced by etoposide in PC 3 cells and the expression of bcl 2 and c myc oncogene.Method TdT mediated x dUTP nick end labeling and flow cytometry were emploied to investigate the apoptosis;c myc and bcl 2 protein were detected immunohisto chemically.Result The findings indicated that the percentage of apoptosis cells was increased and the percentage of positive cells of bcl 2 and c myc expression was decreased with the increasing of concentration and exposure time of VP 16.Conclusion VP 16 can induce the programmed cell death of PC 3 cells and inhibit the expression of bcl 2 and c myc,and the inhibition is dose dependent and time dependent.
