摘要 探讨林蛙油对运动训练小鼠心肌自由基代谢和超微结构的影响,为林蛙油的应用提供实验依据。65只ICR雄性小鼠随机分为安静组、运动组、林蛙油低、中和高剂量组,通过建立小鼠递增游泳训练和游泳力竭模型,测定小鼠力竭游泳时间、心肌SOD活性、MDA含量和小鼠心肌超微结构的变化。林蛙油能延长小鼠力竭游泳时间,与运动组相比,林蛙油中、高剂量组有显著差异(p<0.01)。运动组SOD活性显著降低,与安静组比较,有显著差异(p<0.01);与林蛙油各剂量组比较,中、高剂量组有显著差异(p<0.05)。运动组MDA的含量最高,与安静组比较,运动组有显著异(p<0.01);与运动组比较,林蛙油各剂量组均有显著异(p<0.01)。电镜观察结果表明,小鼠心肌组织超微结构,运动组心肌细胞部分肌丝束上肌节明、暗带结构可辨,部分肌丝束上肌节结构消失,肌浆网扩张;线粒体局部数量增多,呈小圆形,致密状,有的线粒体肿胀,嵴断裂呈空化状。林蛙油组肌丝束上肌节结构较清晰,肌节明、暗带结构清晰,该组未见肌丝溶解、消失的改变;线粒体丰富,呈椭圆形,其嵴隐约可见。林蛙油提高小鼠心肌组织抗氧化酶SOD的活性,减少自由基的生成,对运动造成的心肌组织超微结构的损伤有明显的保护作用,能明显延长小鼠游泳至力竭时间,具有提高机体耐力运动能力。 Explore the impact of Rana oil on trained mice myocardium free radical metabolism and uhrastructure, provided the experimental basis for the application and development of Rana oil. ICR mice were divided to Quiet group, exercise group, dose group. The research aimed to establish the oil Rana mice fed the incremental swimming training and exhaustive swimming model. Changes of mice swimming exhausted time, myocardium SOD activity, MDA content and myocardium uhrastructure were measured. The Rana oil can extend the swimming exhausted time of mice, compared with the trained group, Rana oil of high and medium dose group had a significant difference (p〈0.01). SOD activity in the trained group significantly reduced, compared with the quiet group, had a significant difference (p〈0.01) ; comparison with Rana oil each dose group, the high and medi- um dose group had a significant difference (p〈0.05). The highest MDA content was in trained group, compared with quiet group was significantly different (p〈0.01) ;with Rana oil each dose group were significantly different (p〈0.01). The myocardium uhrastructure observation showed that the exercise group cardiomyocytes part of the muscle tow sarcomere that dark band structure can be identified, and part of the muscle tow sarcomere structure disappeared, sarcoplasmic reticulum expanded;local increase in the number of mitochondria,were small and dense, some mitochondrial welling, eristae rupture was like cavitation. Rana oil group showed a clearer muscle tow sarcomere, the bright and dark band were clearly.There were no changes of myofilament dissolved and disappeared. Rich in mitochondria,oval-shaped ridge looming. Rana oil can significantly improve the function of anti-free radical oxidation of mouse Myocardial tissue, had a damage of the Myocardial tissue caused by the movement, it time, had a good role to improve exercise capacity in mice significant protective effect against ultrastructural can prolong the The mice swimming to exhaustive time, had a good role to improve exercise capacity in mice.
