摘要
目的:通过胚胎期给药,以补气血方为对照,探讨补肾方对胎儿宫内发育迟缓(IUGR)模型胎鼠骨骼肌肌酸激酶(CK)活性的影响,进一步探讨补肾及补气血理论与能量代谢间的关系。方法:将孕鼠随机分为正常对照组、IUGR模型组、左归丸组、右归丸组、五子衍宗丸组、四君子汤组、四物汤组、八珍汤组。除正常对照组外,各组于受孕第8天用酒精给孕鼠灌胃致胎鼠IUGR,于第19天剖检胎鼠,检测胎鼠左前肢肌肉CK活性。结果:与正常对照组比较,IUGR胎鼠骨骼肌CK活性明显降低(P<0.01);与模型组比较,左归丸组、右归丸组、八珍汤组胎鼠CK活性提高,差异有统计学意义(P<0.05,P<0.01)。结论:补肾方及气血双补方胚胎期给药能提高胎鼠CK含量,从能量代谢角度为中医"肾为先天之本,肾主生殖发育"和"补母益子"理论提供了实验依据。
Objective:To exploring the relationship between energy metabolization and traditional Chinese medicine(TCM)theory about reinforcing kidney,supplementing Qi and blood through studying the effect of reinforcing kidney prescription on CK(creatinkinase)activity in skeletal muscle tissue of embryonic mice with intrauterine growth retardation(IUGR).Methods:The pregnant mice were randomly divided into 8 groups,normal control group,IUGR model group,Zuogui pills(ZGW,a prescription for reinforcing kidney Yin)group,Yougui pills(YGW,a prescription for reinforcing kidney Yang)group,Wuzi Yanzong pills(WZYZW,a prescription for reinforcing both of kidney Yang and kidney Yin)group,Sijunzi decoction(SJZD,a prescription for supplementing Qi)group,Siwu decoction(SWD,a prescription for supplementing blood)group,Bazhen decoction(BZD,a prescription for supplementing Qi and blood)group.Mice in normal control group were fed normally.IUGR model mice were induced by feeding alcohol.19 days later,CK activity in skeletal muscle tissue of embryonic mice was detected.Results:Compared with normal control group,CK activity in skeletal muscle tissue of mice in IUGR model group was decreased(P0.01).Compared with model group,CK activity was increased in ZGW group,YGW group and BZD group(P0.05,P0.01).Conclusion:CK activity in embryonic mice could be enhanced through administrating ZGW,YGW,BZD in embryonic period,which may provide an evidence for following TCM theory from the aspect of energy metabolism-"kidney controlling reproduction and upgrowth"、"invigorating the mother to benefit the children".
