白山毛桃根提取物对人红白血病K562细胞作用研究 被引量:4

摘要 目的研究白山毛桃根提取物对人红白血病K562细胞增殖的抑制作用及其可能的作用机制。方法 MTT法检测白山毛桃根提取物对K562细胞增殖抑制的影响;Hoechst33258染色实验观察白山毛桃根提取物诱导K562细胞凋亡情况;流式细胞仪检测该药物诱导K562细胞凋亡的情况。结果白山毛桃根提取物可以抑制K562细胞增殖,且呈时间和浓度依赖性,对K562细胞的半数抑制率(IC_(50))在24 h、48 h、72 h时分别为31、28、10 mg/ml,形态学观察表明白山毛桃根提取物能诱导K562细胞的凋亡。结论白山毛桃根提取物能抑制K562细胞增殖和诱导细胞凋亡。 Objective To investigate the effects of the extractive of radix actnidiae erianthae on the proliferation of human melanoma K562 cells and to further explore its molecular mechanism. Methods The inhibitory rate of cell growth was measured by MTT; the cell apoptosis was detected by fluorochrome of Hoechst33258 and flow cytometry (FCM). Results The extractive of radix actnidiae erianthae had significant activity in inhibiting K562 cells, showing dose dependency. The half-inhibition rate (IC50) on the K562 cell was 31mg/ml, 28mg/ml and 10mg/ml at the time points of 24h, 48h, and 72h, respectively. The morphological change showed that the extract could induce the K562 ceils apoptosis. Conclusion The extractive of radix actn;diae erianthae could significantly inhibit the proliferation; the mechanisms may be related to reducing the K562 cells apoptosis.
