摘要
目的探讨抑郁症患者血浆中miR-155的表达水平及IL-2、IL-6、IFN-γ的浓度变化及意义。方法用实时荧光定量RT-PCR(qRT-PCR)法检测52例抑郁症患者治疗前和治疗8周后血浆miR-155的水平;用ELISA法检测血浆IL-2、IL-6、INF-γ浓度,以40例体检健康者为健康对照。结果抑郁症患者治疗前血浆miR-155水平及IL-2、IL-6、INF-γ浓度明显高于治疗后和健康人对照组(P均<0.01)。治疗前血浆miR-155水平与IL-2、IL-6、INF-γ浓度呈正相关(r分别为0.753,0.639,0.671,P均<0.01)。结论抑郁症患者血浆miR-155的水平较高,且与炎性细胞因子浓度呈正相关,miR-155在抑郁症免疫失衡机制中发挥一定的作用。
Objective To investigate the variation of the expression level of miR-155 and its correlation with proinflammation cytokines IL-2,IL-6 and INF-γ in plasma of the patients with depression.Methods Real-time RT-PCR was used to determine the expression of miR-155 in plasma from 52 cases of depression patients before and after 8 weeks of antidepressant treatment.ELISA was used to measure the levels of IL-2,IL-6 and INF-γ in plasma,and 40 healthy individuals were used as normal controls.Results The expression of miR-155 and levels of IL-2,IL-6 and INF-γ in plasma of depression patients before the treatment were significantly higher than those after treatment and control group(P&lt;0.01).The expression of miR-155 was positively correlated with the levels of IL-2,IL-6 and INF-γ(r=0.753,0.639 and 0.671 respectively,P&lt;0.01).Conclusion The expression of miR-155 in plasma of depression patients before treatment was significantly higher than that in the patients after treatment and healthy control,and correlated with the concentration of proinflammation cytokines,which indicated that miR-155 may play certain role in the mechanism of immunological imbalance of the patients with depression.
