南方红豆杉多糖的含量测定及体外降血糖活性研究 被引量:17

摘要 采用水提醇沉法提取南方红豆杉多糖,硫酸-苯酚法测定多糖含量;采用HepG2细胞和α-葡萄糖苷酶作为体外受体模型,研究南方红豆杉多糖的降血糖效果。结果表明:南方红豆杉药材中多糖的含量为1.75%,无水葡萄糖在1.507~13.563μg/mL内呈良好线性关系,平均回收率97.54%,RSD为1.60%;以二甲双胍和阿卡波糖作为阳性药,南方红豆杉粗多糖对胰岛素抵抗HepG2细胞的葡萄糖消耗具有促进作用,对α-葡萄糖苷酶具有较好的抑制活性,粗多糖抑制活性(IC5038.61mg/L)明显高于阳性对照药(IC501081.41mg/L),且呈现剂量依赖性。 The polysaccharide of Taxus chinensis var. mairei was extracted by hot water, the content of polysaccharide was determined by phenol-sulfuric acid method;the HepG2 cells and alpha-glucosidase were used as receptor model to study the hypoglycemic activity in vitro of Taxus chinensis var.mairei polysaccharide. The results showed that the average content of polysaccharide from the branches and leaves of Taxus chinensis var mairei was 1.75%.When the linear range of anhydrous glucose was 1.507 to 13.563μg/mL, there was a good linear relationship.The average rate of recovery was 97.54%, RSD was 1.60%.Using Metformin and Acarbose as the positive drug,crude polysaccharides from Taxus chinensis var.mairei could improve the glucose consumption of insulin-resistance HepG2 cells, it (IC50 38.61mg/L)showed higher inhibitory activity to alpha-glucosidase than Acarbose( IC5o 1081.41 mg/L).And it showed dose dependent for the inhibitory activity.
