摘要
采用PSA培养基法,从地黄病根上分离得到镰刀菌.通过菌落形态、色素颜色、菌落生长速率、大型分生孢子、小型分生孢子以及产孢梗等形态学观察,初步认为其为层出镰刀菌;采用真菌rDNA ITS区的通用引物对其DNA进行PCR扩增,PCR产物经连接转化并验证后测序,将测序结果登录GenBank进行BLAST分析,检测结果与形态学鉴定结果一致,表明地黄根腐病病原菌JZ1为层出镰刀菌Fusarium proliferatum(其序列在GenBank中的登录号为HQ113948).生物学活性结果表明:层出镰刀菌在15~35℃均可生长,最适生长温度25℃左右;适宜病菌生长的pH值为7.0;其菌丝在蔗糖上生长最好,硝酸钾是其最佳氮源.
In this paper, the pathogen of Rehmannia glutinosa was studied by routine morphological observation and molecular detection. And the biological characteristics of pathogen were also studied. The pathogen was preliminarily identified as Fusarium proliferatum according to its morphology, color and growth rate of the colonies, and morphology of macroconidia, microconidia, chlamydospores, and conidiophores. The fungal rDNA ITS was amplified and sequenced. The sequencing results was accept- ed by GenBank and the accession number is HQl13948. The results of molecular detection and morphologic observation both showed that the pathogens causing root rot of Rehmannia glutinosa is F. proliferatum. The pathogens could grow at 15 to 35 ℃ and the optimum temperature was 25 ℃, the optimum pH value was 7.0. The optimum carbon source and nitrogen source were sucrose and potassium nitrate respectively.
