3-溴丙酮酸治疗人胃癌细胞动物模型的实验研究

摘要 目的:探讨3-溴丙酮酸(3-Bromopyruvic acid;3-BrPA)对人胃癌细胞株SGC-7901裸鼠皮下种植瘤的抑制作用。方法:建立人胃癌裸鼠皮下种植瘤模型,将成瘤后的裸鼠随机分为实验组:3-BrPA低、中、高剂量组,PBS(磷酸盐缓冲液)阴性对照组1(pH7.4)、对照组2(pH6.8~7.8)及阳性对照5-FU组。连续4周每天瘤体周围皮下注射给药,停药24h后取下瘤体;原位末端标记法(TUNEL法)检测肿瘤细胞凋亡指数,透射电镜下观察肿瘤组织细胞形态及结构变化;计算肿瘤的体积抑瘤率。结果:3-BrPA给药组的瘤体积分别与2个PBS组比较均有意义(P<0.05),其中高剂量组与5-FU组差异无统计学意义(P>0.05)。透射电镜观察到3-BrPA低、中、高组及5-FU组裸鼠的肿瘤组织细胞均出现典型的凋亡特征。TUNEL法检测凋亡指数显示:实验组凋亡指数分别与2个阴性对照组比较均有意义(P<0.05),其中5-FU组凋亡指数与中剂量组差异无统计学意义(P>0.05),而与高剂量组差异有统计学意义(P<0.05)。结论:3-溴丙酮酸在体内对人胃癌细胞SGC-7901种植瘤有较强的抑制作用。 Objective To investigate the effects of 3-bromopyruvic acid (3-BrPA) on the growth of implanted human gastric cancer cell line SGC-7901 in nude mice. Methods The subcutaneous implanting tumor models of human gastric carcinoma in nude mice were established, and then were divided into 6 groups: low, median, and high dose 3-BrPA groups, PBS control group 1 (pH 7.4), PBS control group 2 (pH 6.8 - 7.8), and 5-fu control group. Mice in each group received subcutaneous injection for 4 weeks. The tumor inhibition rate was calculated to evaluate the anti-tumor effect. The apoptosis rate of gastric carcinoma cells was measured by TUNEL method. The tumor cells structure were detected by TEM. Results As compared to those in PBS control group 1 and 2, in 3- BrPA and 5-FU groups, the growth of implanting tumor was obviously inhibited (P 〈 0.05 ), the classic apoptosis feature could be observed, and the apoptosis index was significantly different (P 〈 0.05). Conclusion 3-BrPA can inhibit SGC-7901 growth in mice.
