系统性红斑狼疮患者外周血淋巴细胞TGFβR2和信号传导分子Smad2的表达

Expressions of TGFβR2 and Smad2 in peripheral lymphocytes in patients with systemic lupus erythematosus

摘要 目的:检测系统性红斑狼疮(SLE)患者外周血淋巴细胞转化生长因子β受体Ⅱ(TGFβR2)和信号传导分子Smad2 mRNA表达,探索其在SLE发病中的作用。方法:采用RT-PCR半定量方法,对48例SLE、20例正常对照和12例RA外周血淋巴细胞TGFβR2和Smad2 mRNA表达进行检测,并结合临床资料进行分析。结果:活动组SLE(0.377±0.365)外周血淋巴细胞TGFβR2mRNA表达明显低于正常对照组(0.829±0.311)和稳定组SLE(0.799±0.34)(P<0.01);SLE患者TGFβR2表达与SLEDAI评分和抗ds-DNA抗体负相关(r=-0.556和r=-0.49,P<0.05),与补体C3正相关(r=0.62,P<0.01)。活动组SLE外周血淋巴细胞Smad2 mRNA表达低于稳定组(0.578±0.329vs0.801±0.29)(P<0.05),但与正常对照组(0.733±0.323)无差别(P=0.144);SLE患者Smad2表达与SLEDAI评分负相关(r=-0.42,P<0.05)。结论:活动SLE患者TGFβR2和Smad2表达减少,可能使TGFβ对淋巴细胞的免疫抑制作用减弱,提示TGFβR2和Smad2异常表达可能参与SLE发病,并对判断病情活动等有一定意义。 Objective To investigate the expressions of TGFβR2 and Smad2 and their clinical significances in peripheral lymphocytes in patients with systemic lupus erythematosus (SLE). Methods Expressions of TGFβR2 and Smad2 in peripheral lymphoeytes in 48 patients with SLE, 20 normal controls, and 12 patients with rheumatoid arthritis (RA) were measured by semi-quantitative RT-PCR, and the clinical data were analyzed accordingly. Results Expression of TGFβR2 mRNA in active SLE patients (0.377 ± 0.365) was significantly lower than those in inactive SLE patients (0.799 ± 0.34) and normal controls (0.829 ± 0.311 ) (both P 〈 0.01 ). The level of TGFβR2 in SLE patients was inversely correlated with SLEDAI and anti-ds-DNA antibody level (r = -0.556, r = -0.49, P 〈 0.05), and was positively correlated with C3 level (r = 0.62, P 〈 0.01). Expression of Smad2 mRNA was lower in active SLE patients than those in inactive SLE patients (0.578 ±0.329 vs. 0.801 ± 0.29) (P 〈 0.05). The level of Smad2 was negatively correlated with SLEDAI (r = -0.42, P 〈 0.05). Conclusion Expressions of TGFβR2 and Smad2 were decreased in active SLE patients, which indicated that they may play certain roles in the pathogenesis of SLE.
