电针丰隆穴对高脂血症大鼠主动脉弓ABCA1mRNA表达和血液流变学的影响 被引量:1

Effects of Electroacupuncture at Fenglong(ST 40) on ABCA1 mRNA Expression and Hemorheology in Rats with Hyperlipidemia

摘要 目的:探讨电针丰隆穴对高脂血症的治疗作用及其作用机制。方法:36只健康SD大鼠随机分为空白组、模型组、电针组,每组12只。采用高脂饮食饲料饲喂法建立高脂血症大鼠模型。造模28天后,电针组开始电针丰隆穴治疗。治疗结束后,检测各组大鼠血脂及全血黏度、血浆黏度、红细胞聚集指数,并用原位杂交法检测各组大鼠主动脉弓组织中ABCA1mRNA表达水平。结果:与空白组相比,模型组大鼠TC、LDL-C水平显著上升(P<0.01),主动脉弓ATP结合盒转运子A1(ATP binding cassette transporter A1,ABCA1)mRNA表达降低(P<0.01),全血黏度、血浆黏度及红细胞聚集指数明显升高(P<0.01,P<0.05);与模型组比较,电针组大鼠TC、LDL-C水平显著降低(P<0.01),主动脉弓AB-CA1mRNA表达升高(P<0.05),全血黏度、血浆黏度及红细胞聚集指数明显降低(P<0.01,P<0.05)。结论:电针丰隆穴有良好的降脂及防治AS的作用,其机制与上调ABCA1表达以及改善血液流变性有关。 Objective : To investigate the effect and mechanism of electro - acupuncture (EA) at Fenglong (ST40) on blood lipids in hyperlipidemia (HLP) rats. Methods:30 healthy SD rats were randomized into a normal control group, a model group, an EA group, 12 rats in each group. The normal group was fed with basal forage, and the other 4 groups were fed with high fat forage each day to establish hyperlipidemia rat model. 28 days after modeling,the EA group started to do electro -acupuncture at Fenglong (ST 40). At the end of the treatment ,the lipids!161ood viscosity including high, medium and low shearing rates and plasma viscosity erythrocyte aggregation index( A1), were assayed by using a full - automatic hemorheo - lometer and expression of ATP binding cassette transporter A1 ( ABCA1 ) mRNA in aortic arch tissue was assayed by in situ hybridization (ISH). Results : The contents of TC and LDL - C were apparently increased and the expression of ABCA1 mRNA was significantly decreased in the model group (P 〈0. 01 ) ;compared with the model group,levels of TC and LDL - C were significantly decreased (P 〈0. 01 ) and the expression of ABCA1 mRNA was significantly increased in the EA group( P 〈 0. 05 ). Regarding the parameters of hemorheology, compared to the normal group, the high, medium and low shear rates and erythrecyte AI in the model group were significant (P 〈 0.01, P 〈 0.05 ) ;compared to the model group, the high, medium and low shear rates and erythrocyte AI in the EA group were down -regulated significantly( P 〈 0, 01 ,P 〈 0. 05 ). Conclusion :Electro - acupuncture at the acupoint Fenglong can function in lowering lipid and preventing atherosclerosiS by improving the expression of ABCA1 mRNA and facilitating cerebral blood circulation.
