摘要
研究油菜籽清油的提取工艺并测定了清油的理化性质和脂肪酸组成,通过对碱性蛋白酶Alca-lase 2.4L、Protex 6L和中性蛋白酶Protex 7L的筛选,发现碱性蛋白酶Alcalase 2.4L的清油出油效率最高。选用碱性蛋白酶Alcalase 2.4L,在单因素试验的基础上,通过响应曲面优化蛋白酶酶解提取菜籽清油工艺,得到最佳参数为:加酶量1.50%,酶解温度60℃,酶解时间100 min,酶解pH 9.0,此时菜籽清油出油效率为62.31%。且本研究所制得的清油理化指标均符合国际标准,其中不饱和脂肪酸高达92.45%,是很好的食用性油脂。
Study on technology of free oil extraction from rapeseed, through the screening of protease Alcalase 2.4L, Protex 6L and Protex 7L,we found that the effect of protease Alcalase 2.4L is best, so we choose it. On the basis of single factor experiment, through the response surface optimization protease enzyme solution extraction rape- seed oil process, we got the best parameters as follows:enzyme addition 1.50%, hydrolysis temperature 60 ℃, hydrolysis time 100 min, enzyme solution pH 9.0, this time the free oil of rapeseed yield is 62.31%. Extraordinary, free oil is a good oil whose physical and chemical indexes accords with the international standards,the unsaturated fatty acid as high as 92.45 %.
